Experimental Study Of The Protective Effect Of Bellidifolin On Cerebral Ischemia Injury

Background and purpose:Cerebral ischemia injury accounts for 60-70% cerebral vascular diseases.No medicine is available in cerebral ischemia injury.Bellidifolin is extracted from "Artemisia Capillaris Thurb" which has extensive pharmacological effect.The present study is to explore the protective effect of bellidifolin on cerebral ischemia injury and its possible protective mechanism.Methods and Results:1. Effect of bellidifolin on duration of gasping after the complete cerebral ischemia induced by decapitation in mice75 mice were arranged in 5 groups which were NS group,nimodipine group and 50mg/Kg 75mg/Kg, 100mg/Kg bellidifolin group.Complete cerebral ischemia model was induced by decapitation in mice 30 minutes after administration.The duration of gasping was recorded.Compared with NS group ,50mg/Kg bellidifolin, 75mg/Kg bellidifolin and 100 mg/Kg bellidifolin groups could prolong the time significantly by 7.78%(0.010.05) , 8.77%(P<0.01) , 5.26%(P=0.05) compared with NS group;nimodipine group could decrease the brain index by 4.91%(P<0.01) compared with NS group;50mg/Kg bellidifolin group had a significant difference compared with nimodipine group (0.010.05) , 15.41%(P<0.01) , 19.46%(P>0.05);nimodipine group could increase brain SOD activity by 21.39%(P<0.01).(3).Compared with NS group , bellidifolin PO 25 , 50, 75mg/Kg group could decrease brain MDA content by 5.31%(P>0.05) , 33.46%(P<0.01) , 10.93%(P>0.05);nimodipine group could decrease brain MDA content by 22.53%(P<0.01).(4).Compared withNS group , bellidifolin PO 25, 50, 75mg/Kg group could increase the chromaticity which accounts for the content of brain Glu by 4.36%(P>0.05), 10.38%(0.010.05) , 14.01%(P<0.01) , 13.80%(P<0.01);nimodipine has not a significant increase which was 3.82%(P>0.05).(6).Pathological inspection results showed :the cells of sham operation group were regular,construction was intact,cell arrangement was in order;but the cells of NS group had changed:some cells turned triangle,nuclei were densed,a broader gap appeared around the nervous cells;the brain injury of bellidifolin PO 25 , 50, 75mg/Kg group was great improved.Conclusion:These results suggest that bellidifolin play a protective role in cerebral ischemia injury and anti-oxidation , anti-excitement virulence mechanisms take part in the protective process.
