The Effect Of Cyclosporine A On The Trophoblast Cell Secretion Of Interleukin-10 (IL-10) And Interferon-gamma (IFN-γ) In Early Pregnancy In Vitro Culture

objective : To investigate the effect of cyclosporine A (CsA) on the trophoblast cell secretion of interleukin-10 (IL-10) and interferon-gamma(IFN- γ ) in early pregnancy in vitro culture. Method: Isolate and purificate 6-8weeks of gestation trophoblasts. Digest it by trypsm , culture it in vitro and treat it with CsA 0 μ mol/L, 0.1 μ mol/L, 1.0μ mol/L and 10.0μ mol/L respectively. Assay the IL-10 and IFN-γ by immunohistochemical staining and computerized image analysis techonologe. the IL-10 and IFN- Y mRNA expressions were determined by RT-PCR. Results(l) The IL-10 and IFN- γ were well manifested on the cytoplasm and membrane of trophoblast cell. (2)CsA have obviously increased the secretion of protein IL-10 in trophoblast cell at 0.1 and 1 μ mol/L, and there was no statistical significance among the 2 concentrations of CsA (P>0.05). However, the secretion of IL-10 was not affected when CsA was either of 0 μ mol/L or of 10μ mol/L and there was no statistical significance among the 2 concentrations(P>0.05). IFN- Y protein manifested in the trophoblast membrane of cell in the 4 CsA concentrations were almost the same and there were no statistical significance among the 4 concentrations (P>0.05). thebalance of Thl/Th2 deviate towards Th2 which benefits to pregnancy.(3) mRNA expressions are same as the results of immunohistochemical staining.Conclusion: CsA can well control the secretion of cytokine of the trophoblast cell when CsA is in a range of suitable concentrations.
