As positiive controlling factor of cell cycle, the oncogene cyclinE is a key regulatory gene in its entry into S phase. While as negative controlling factor of cell cycle, p27kip1 is a member of the CEP/KIP family of cyclin-dependent kinase (CDK) inhibitors,and regulates cell cycle in various kinds of ways . p27kip1 preferentially binds cyclinE/CDK2 complexes,inhibits their CDK activity,and thus obstructs entry into the S-phase, resulting in G1 phase arrest. The discoordiation of cyclinE and p27 kip1 will lead to the proliferation and malignity of cell. The objective of this study is : (1) to observe the expression of p27 Kip1 and cyclinE in gastric carcinoma of three different pathologic types (2) to investigate relationship between p27 Kip1 and cyclinE in gastric carcinoma (3) to investigate the relationship between Hp and expression of p27 Kip1 /cyclinE in gastric carcinoma.Methods: We detected the expression of p27 kip1 and cyclinE in 59 gastric cancer samples by immuno-histochemical staining technique (SP method), we can analyze the different expression of p27 Kip1 and cyclinE in different pathologic types. In this study , Hp was examined by Giemsa staining and rapid urease test.,We classified gastric cancer into three pathological types by HE staining.Results: (1 )The expression of p27 Kip1 and cyclinE in normal gastric mucosa cells and gastric carcinoma: The positive expression of p27 Kip1 in normal gastric mucosa cells and gastric carcinoma was 84.22%(3 of 19), 47.46% (28 of 59) respectively. There were statical significance between normal gastric mucosa and stomach carcinoma.(X2=7.89 p<0.05).There were only 2 samples cyclinE positive expression in normal gastric mucosa and positive expression rate of cyclinE was 55.93%(33 of 59). There was obvious statical significance between normal gastric mucosa and stomach carcinoma.( X2=11.97 p<0.05) (2) The expression of p27Kip1 and cyclinE in different pathological type of gastric cancer: The positive expression of p27 Kip1 in high differentiation adenocarcinoma , low differentiation adenocarcinoma and stomach mucoid carcinoma was 68.42%(14 of 19), 37.5% (9 of 24), 31.25% (5 of 16) respectively. There was ststical significance between high differentation adenocarcinoma and low differentitation,stomach mucoid carcinoma,( X2=5.58 p<0.05 X2=6.3 p<0.05 respectively) whereas ,there was no significance between low differentitation adenocarcinoma and stomach mucoid carcinoma ( X2=0.16 p>0.05) .The positive expression of cyclinE in stomach mucoid carcinoma ,low differentitation adenocarcinoma and high differentitation adenocarcinoma was 68.75% (11 of 16), 66.67% (16 of 24) and 31.58% (6 of 19), respectively. The expression of cyclinE was obvious higher in stomach low differentitation adenocarcinoma and stomach mucoid carcinoma than in stomach high differentiation adenocarcinoma (X2=4.8 p<0.05 X2=5.23 p<0.05 respectively), whereas there was no statical significance between low differentitation adenocarcinoma and stomach carcinoma (X2=0.02 p>0.05). (3) The relationship between the expression of p27 Kip1 and cyclinE in gastric cancer: The positive expression of cyclinE in the negative expression of p27 Kip1 is higher than that in the positive expression . A negative correlation was observed between the expression p27Kip1 and cyclinE (p<0.05 ). (4) The influence of Hp infection associated gastric cancer on p27 Kip1 and cyclinE: The expression of p27 Kip1 between Hp positive and Hp negative gastric carcinoma were 48.15% and 46.88% respectively.
