Since water pollution is becoming more and more serious in the world, varieties of water products come forth to serve as drinking water, among which purified-water is the most significant and covers the majority of the world market. Although drinking purified-water can avoid the harm resulting from organic or inorganic substances in ordinary drinking water, as a kind of low mineral water, purified-water hardly has any mineral elements, such as calcium (Ca2+), magnesium (Mg2+) and so on, which are essential to human health. Many reports in home and abroad have indicated that drinking low mineral water for a long time increase the incidence of cardiovascular diseases, especially atherosclerosis (AS). Our previous study also have showed that the rats drinking purified-water from antenatal to 20 weeks postnatal have higher blood lipids (TC, TG) and lower concentration of Mg2+ in the serum among total 16 mineral elements determined in the experiment. With the development of our society and living condition, people especially those living in urban area generally have a life of eating high-fat diet and drinking purified-water. High-fat diet can cause hyperlipidemia in animals and human beings, which is regarded as the major risk factor for AS. Cardiovascular and cerebrovascular diseases caused by AS are seriously harm to human health.Mg2+, an important endocellular positive ion besides K+, has many important physiological and biochemical effects in human body. In a word, it takes part in any reactions ATP attending. Results from animal experiments and clinical studies suggest that Mg2+ can decrease the blood contents of lipids, cure or prevent cardiovascular diseases. Mg2+ plays an important role in the development of AS. Mg2+ deficiency leads to AS, while AS progress can be prevented when oral administration of magnesia mixture is taken.But most previous studies on low mineral water or high-fat diet focused on single factor, and ignored the combined effect. With the development of living standard, people pay more and more attention to combined action of multiple factors. On the basis of our previous studies, considering the actual society situation, we simulated the modern life style to feed the rats with purified-water and high fat diet at the same time to explore the effect of the two factors in the development of hyperlipidemia and AS.Methods: A total of 150 female Wistar rats were employed and randomly equally divided into five groups, groupâ… fed with tap-water and basic feedstuff (TW), group â…¡ with purified-water and basic feedstuff (PW), group â…¢ with purified-water supplemented with 0.5g/L MgSO4 and basic feedstuff (PW+Mg), group â…£ with purified-water and high-fat diet (PW+HFD), and groupâ…¤with 0.5 g/L MgSO4 purified-water and high fat diet (PW+ HFD +Mg). The high fat diet is composed of 95% basic feedstuff, 5% lard and 1% cholesterol, which are half of the feedstuff inducing experimental rat models of AS. All rats were fed with the mentioned-above waters and the feedstuffs respectively for five months. Three rats were bred in one cage, and are free for water and food whose amounts were recorded every time. Body weight and sebum thickness of the rats in every group were measured at the end of every month, and then after being fasted for 12 h, their blood samples were collected from tail vein for the dynamic determination of serum concentrations of total cholesterol(TC) and triglycerol(TG). Five months later, after the blood samples were obtained through femoral artery of every rat, the rats were then killed and their vessel (aortic arch), heart (frontal wall of left ventricle), liver (lateral middle part of left lobe), and spleen (middle part) were taken and fixed in 4%formaldehydum followed by embedded in paraffin 72 h later. The blood concentrations of TC, TG, high density lipoprotein (HDL-C), and low density lipoprotein (LDL-C) were determined by enzyme link chromatometry, of Ca2+, Mg2+, malonic ldehyde (MDA), vitamin E (VE) and nitric oxide (NO) by chemical chromotometry, and endothelin...
