To make clear the effect of VEGF and NO on the mechanism of pregnancy-induced hypertension syndrome (PIH), and to investigate the potential curative mechanism of improving the ischemic and anoxic state of the placenta in HuangQiDanShen compound prescription based on Qi-tonifying and bloodstasis-resolving therapy (YiQiHuaYu) in TCM.Methods: NOS blocker L-NANE was used to make PIH animal model on the 10th pregnant day. The active components of astragalus root and red sage root were extracted and made into injection (named "QIDAN injection"), and nitroglycerin and the QIDAN injection were administered synchronously. Blood pressure, urine protein, the weight, the weight of brain, the weight of liver and spleen of infantile rats, the weight of placenta, the mortality rate of fetus, the level of NO in plasma ,the level of VEGF in placenta, and so on were observed.Results: By inhibiting the synthesis of NO hypertension, albuminuria and some other PIH-related symptoms occurred in the pregnant rats, and the rats of intrauterine growth retardation (IUGR) and fetal death in the uterus increased obviously. QIDAN injection improved all measured growth parameters of the fetus. The incidence of IUGR and fetal death in uterus decreased remarkably in the treatment group. QIDAN injection decreased the levels of urine protein and blood pressure in pregnant model rats. QIDAN injection improved uterine and placental circulation by regulating the endothelial cell's function of secreting NO. QIDAN injection adjusted the levels of VEGF at placenta to increase the placental blood circulation.Conclusions: The results have shown that: By inhibiting the synthesis NO hypertension can establish PIH animal model. VEGF and NO synthetical deficiency is an important factor or a process in the pathogenesis of PIH. QIDAN injection based on the YiQiHuaYu therapy can improve the uterine and placental circulation by affecting the levels of NO and VEGF, promoting the trophoblastic cellular inflitration and accelerating the process of placental revascularization.
