| Objective To study the relationship between the apolipoprotein E(ApoE) gene polymorphism and Alzheimer disease( AD) and the effects of ApoE gene polymorphism on the metabolism of serum lipids in AD. Methods The polymorphism of the ApoE gene was detected using rapid-cycle polymerase chain reaction(PCR) and fluorescence resonance energy transfer(FRET) technique for 25 patients with AD and 59 normal controls. The serum levels of ApoE, triglyceride(TG), total cholesterol (TC) were measured.Results The most frequent genotype was 3/ 3 ,but 2/ 2 was not seen in the 2 groups. The estimated ApoE allele frequencies (%)in AD group were 10, 62, 28 for the 2, 3, 4 alleles respectively; and 7.63, 78.81 , 13.56 respectively in the control group.The frequencies of the ApoE 4 in AD group (28%) were significantly higher than those in the controls ( 13.56%) (p<0.05). 12 ApoE 4 carriers in AD group of 25 and 13 carriers in control group of 59. ApoE 4 has significant relation to the occurrence of AD(p<0.05), RR=2.2. The serum concentrations of [ApoE (39 5) mg/L, TG (1.35 0.31 ) mmol/L, TC (4.4 0.5)mmol/L] in AD were significantlylower than those[(43 8 )mg/L, (1.55 0.22)mmol/L, (4.8 0.6)mmol/Lrespectively] in the controls (p<0.05).Conclusions ApoE 4 allele has definite relation to the occurrence of AD, it is a risk facter for the occurrence of AD. There is abnormal metabolism of serum lipids in AD. |
