PrefaceGallbladder cholesterol cholelithiasis is an exceptionally common digestive disease. Epidemiologic studies indicate that cholesterol cholelithiasis is related to kindred inheritance. The formation of cholesterol gallstone probably results from the action of complex environment and unknown poly gene. Gallbladder cholesterol cholelithiasis represents a failure of biliary cholesterol homeostasis in which the physicalcherm'cal balance of cholesterol solubility in bile is disturbed, the pathophysiologic defect in both mice and humans is unphysiologic cholesterol supersaturation of gallbladder bile. The reason may be as follows : first , cholesterol of liver is secreted overfull. Second, secretion of bile salt and phosphati-dylcholine is decreased. It is reported that MRP2 is related with secretion of bile salt. In this study ,we intend to discuss MRP2 genes action in Gallbladder cholesterol gallstone formation by comparing; MRP2 genes expression between Gallbladder cholesterol cholelithiasis patients and the man without cholelithiasis with the aid of RT - PCR.Patients and MethodsTissue specimens. 14 liver tissue specimens were taken from patients with Gallbladder cholesterol cholelithiasis, which are the trial group. The control group's tissue specimens were taken from 5 patient with trauma hepatorrhexis, which have been proved without cholelithiasis.RNA isolation and cDNA synthesis. Total UNA was isolated from the liver specimens by using Trizol reagent. cDNA were synthesized from total RNA by u-sing Reverse Transcription System reagent.Reverse Transcription - Polymerase Chain Reaction. Polymerase Chain Re-action (PCR) conditions were firstly at 94 C for 5 minutes, secondly, at 94 C for 30 seconds, at 60 C for 40 seconds, at 72C for 1 minute, which is for 30 cycles. Finally, at 72C for 5 minutes. The PCR productions were identified by polypropylene gelatin electrophoresis.ResultsThe MRP2 mRNA's expression in the liver specimens of patients with gallstone was obviously lower than that of patients without gallstone. The difference between trial group and control group has marked significance. ( p <0.01 )DisscussionMRP2 gene belongs to multidurg resitance protain transporters gene family binding ATP. A lot of study indicate that MRP2 in human and mrp2 in mice were almost located in the canalicular membranes of the liver. MRP2 transports divalent bile salt. In this study, we find that The MRP2 mRNAs expression in the liver specimens of patients with gallstone was obviously lower than that of patients without gallstone by Semi -quantitative analysis of MRP2 gene mRNA expression , which prompt that MRP2 may affect the gallstone formation. As MRP2 was bile salt transporter, if the MRP2 mRNAs expression was decreased, the MRP2 will be cut down and the bile salt in the bile will be reduced. However, bile salt is an important element in the bile, which enhance cholesterol dissolve and prevent gallstone formation. So the bile salt decrease will lead to debasement of cholesterol dissolve and gallstone formation.Conclusion1. the MRP2 mRNA expression was decreased in the liver of patients with Gallbladder cholesterol cholelithiasis.2. the MRP2 gene may affect the gallstone formation.
