Objective : To explore the effects of interleukin(IL)-18 on Th1/Th2 cells activity in asthma and the effects of different drugs on it . Methods: Venous blood from 30 asthma patients and 15 health subjects were isolated by density centrifugation with Ficoll-Hypaque, then plasma samples of asthma and health group were collected. ELISA method was used to determinethe concentrations of IL-18, 1L-12 and 1L-13 in plasma. Meantime PBMC from asthma group were obtained and divided into three groups: control group, dexanethasoe group and IFN- group, they were cultured together, the concentrations of three cytokines in cultured PBMC were determined with ELISA. Results: The levels of TL-18 and IL-L3 in asthma group were remarkably higher than that of healthy ones, but IL-L2 levels was much lower than normal subjects in plasma. There are significant difference in statistics(p<0.01). After dexanethasoe intervention in cultured PBMC, the levels of three cytokines were much lower than that of control group(p<0. 05) ;After IFN- Y intervention in cultured PBMC , IL-12 was remarkably higher than that of control group(p<0.05), but the levels of IL-18 and IL-13 had not significant difference with control group (p>0.05). Conclusion: (1) IL-18 is involved in the asthma process and plays an important role in affecting Th1/Th2 cells activity and cytokine production. (2) The combination of dexanethasoe with IFN- might be the best method to cure asthma.
