Objective: To investigate the expression and role of vascular endothelial growthfactor (VEGF), von Willebrand Factor (vWF), very late antigen alfa-2 (VLA-2a) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in the angiogenesis and invasion of xenografted human glioma in different growth periods. To offer experimental thereunder for selecting therapy-target of glioma from the aspect of tumor stroma.Methods: With malignant human glioma cell-line SHG-44 and SHG-44xenografted nude mice model, the immunocytochemical and immunohistochemical assays (monoclone antibodies against VEGF, VLA-2a, TIMP-2 and vWF serve as primaryantibodies, detecting by S-P ) were done in vivo and in vitro. Cell transplanted glioma (n=6) and normal human brain tissue (n=6) were used as controll groups for tissuetransplanted glioma (n=30) . The data of micro-vascular density (MVD), necrosis index,glioma weight, grafting time and pathological index of VEGF, VLA-2a and TIMP-2 wereanalyzed statistically.Results: The expression rates of VEGF, VLA-2a, TIMP-2 and vWF was 100%in xenografted glioma samples. Most of tumor cells and some endothelial cells expressed protein of VEGF, VLA-2a and TIMP-2 with different expression in different regions of tumor tissue and/or different growth periods. Most of endothelial cells and a few tumor cells expressed vWF protein.
