| Objective: To study the activity of toremifene(TOR) and its synergistic effect with cisplatin(DDP) on Gejiu human lung adenocarcinoma cell line GLC-82 , Which might be expected to provide a new therapy mode for the clinical management of lung cancer. Methods: The cytotoxic effects of these agents on GeJiu human lung cancer cell line GLC-82 were measured by a tetrazolium-based colorimetric assay (MTT assay); Expression levels of P21 and P53 were determined by Western blotting. Results: TOR inhibited the growth of GLC-82 cell .The TOR (>5umol/L) significantly enchanced chemosensitivity of DDP .TOR enhanced the antitumor activity of DDP. And expression of P21 and PS3 were increased.Conclusion: TOR combined with DDP shows significant synergistic anti-tumor effect on GLC-82 cells. The mechanism of effect could be related with change of phase in cell cycles which were blocked by DDP, and could be associated with PS3 and P21 induction and may involve a P53-independent path way. |
