| Objective: Short tandem repeats (STR) which are discorvered recently are DNA sequences with length polymorphisms in human genome. The core sequence which is consisted of 2-6 base pair is lined up repeatedly in length. The number of repeats is 15-30 time and the length polymorphisms are caused by the variety of copy in core sequence. It is belieaved that there is a STR locus in each 6-10kb. STR loci distribute extensively and have higher information, polymorphism, and their heredities are based on Mondel codominant inheritance, therefore, they have become the ideal polymorphic markers for genome mapping, genetic linkage analysis, individual identification, paternity test and estimation of organic transplantation.The variations of STR provide vast genetic markers which can be used to describe genetic structure of population and extensively used in anthropologic research. Henan province locates in central area of china and has large population, so the genetic character of Han population is agood representative in northern population of china. Using multi-PCR amplification of fifteen STR loci in Henan Han population, we try to get popular genetic data of these loci in Henan Han populaton.Materials and Methods: The DNA samples of 242 Henan Han individuals who required paternity test were extracted by classic SDS-protein K-phenol-chloroform method. After fifteen SIR loci ( CSF1P. TPOX. THO. F13AO. FESFPS. vWA. D16S539. D7S820. D13S317. D12S39. D5S8. D18S5. PAH. D8S1179. D3S1358) had been amplified by multi-PCR amplification, electrophoresis on 6% denatured polyacrylamid gel. The gels were fixed, silver-dyed and coloured until the bands were clear, then individual genotypes were distinguished through the comparison between the amplified bands and the STR characteristic allelic ladder. Weather STR locus ' genotypes accorded with the Hardy- Weinberg equilibrium was determinded by x2 test . The observed heterozygosities(H), expectative heterozygosities(He), power of discrimination(DP), polymorphism information content(PIC), power of exclusion(PE) and matching propability(PM) were calculated by Power Stats software.Result: Eight alleles and thirty genotypes were observed at CSF1PO locus, as well as eight alleles and eighteen genotypes at TPOX locus, seven alleles and fifteen genotypes at TH01 locus, six alleles and fourteen genotypes at F13A01 locus, seven alleles and eighteen genotypes at FESFPS locus, eight alleles and thirty genotypes at vWA locus, nine alleles and twenty-eight genotypes at D16S539, nine alleles andtwenty-six genotypes at D7S820, six alleles and fourteen genotypes at D13S317 locus, nine alleles and twenty-six genotypes at D12S391, eight alleles and twenty- two genotypes at D5S818, eleven alleles and fouty-seven genotypes at D18S51, seven alleles and twenty genotypes at PAHI3, nine alleles and thirty-three genotypes at D8S1179, seven alleles and sixteen genotypes at D3S1358. DP for the fifteen loci is 0.88, 0.824, 0.824, 0.842, 0.863, 0.933, 0.944, 0.926, 0.936, 0.934, 0.911, 0.955, 0.869, 0.952, 0.856 respectively, and PIC is 0.71, 0.59, 0.60,0.62,0.65,0.79, 0.80, 0.76, 0.78, 0.79, 0.74, 0.83, 0.68, 0.82, 0.66 respectively. The combined DP is 0.99999999999999997, the cumulative PE is 0.999915 and the cumulative PM is 3.422 X 10-16.The x2 test of gene frequency in every allele of D7S820 locus among the Henan Han population, Chengdu Han population, Africa-Americans, Caucasian-Americans and Hispanic-Americans indicates that there is significant difference among main races, expect between the Henan Han and Chengdu Han population.In 304 paternity test cases, there are thirty-six cases which exclude paternity (for over four loci are mis-matching). Only in one case among the twenty-three loci, a locus of the child did not accord with his father's, speculating that it might be a mono-locus mutation.
