We could stop the development of tumor probably if the previou hormones were resisted.Mifepristone is the antagonist of progestogen and glucocorticoid receptor.lt is manifest that Mifepristone has selectivity inhibition potentially to the formation and growth of some human tumors.And it has already displayed satisfactory prospect of application on hormone dependence tumor just as hysteromyoma mammary cancer and carcinoma of prostate in clinical therapy.survivin is a new anti-apoptosis gene and it's one of the inhibitor of apoptosis protein.Because of the distinctive structure,special tissul distribution and obviouse anti-apoptosis action,survivin is interested by the scholar of tumor. But it's not mature about the role of Mifepristone to ovarian carcinoma and the regulation to survivin.The article's objective is to investigate the growth influence of ovarian carcinoma cell effected byMifepristone and its mechanism.Objective: To study the effect of mifepristone on cell apoptisis of human ovarian carcinoma cell lines HO8910 in vitro and the effect of survivin gene on cell apoptises.Method: The cells of HO8910 were cultured.In the same growth term,mifepristone of different dosage were added.and the cells growth station was observed. The antiproliferation effects of four different concentrations of mifepristone(20,10,5,2.5 u mol/L)on HO8910 cell line were determined with MTT assay.Flow cytometry techniques was used to to determine cell apoptosis under the action of mifepristone. And expression of survivin gene was evaluated by reverse transcription polymerase chain reaction (RT-PCR).Result: Four different condensations dose-dependently inhibited the proliferation of HO8910 cells.the inhibition by mifepristone plateaued at 20 y. mol./L(72.80%) It has a significant difference compared with control group (P<0.005) .The result of Flow cytometry techniques is showed that the apoptosis rate are 68.49% 12.63% and 5.80%;The content of survivin mRNA is more than that effected by Mifepristone. It has a significant difference compared with control group (P<0.005) .Conclutions: It is suggested that mifepristone can inhibit the proliferation of epithelial ovarian cancer cells in vitro and there is an accordant dose-time-response relationship.Its anticancer effect seems to be due to induction of apoptosis which maybe a result of dowm-regulation of the anti-apoptotic gene survivin. | | Keywords/Search Tags: | mifepristone, Ovarian neoplasms, survivin, Apoptosis | PDF Full Text Request | Related items |
