| Objective: To investigate the value of RASSF1A promoter methylation and cyclinD1 expression in lung cancer diagnosis, and analyse the relativity between RASSF1A promoter methylation and the expression of cyclin D1。Methods: Methylation-specific PCR(MSP)was performed for detecting the methylation status of RASSF1A promoters in 33 cases of lung cancer and 10 paracancerous normal lung tissue samples.Immunohistochemiscal S-P method was used to detect the expression of cyclin D1 in all the samples above.Results: 18 lung cancer cases (54.5%)showed that their RASSF1A gene were methylated,and cyclin D1 expressed in 16 out of 33 (48.5%) lung cancer samples. but no RASSF1A gene methylation were detected in the 10 paracancerous normal lung tissue samples, and immunohistochemical staining of these samples for cyclinD1 were negative. either RASSF1A methylation or cyclin D1 expression had no relationship with gender, age and histology type .The expression of cyclin D1 had correlation with differentiation of lung cancer(r=0.418, P<0.05), but No relationship was found between RASSF1A gene methylation and cyclin D1 expression in lung cancer. Conclusion: RASSF1A methylation and overexpression of cyclin D1 play important roles in carcinogenesis of lung. The expression of cyclin D1 has correlation with the differentiation of lung cancer, and can be used to judge the degree of malignancy of lung cancer. |
