Preparation And Characterization Of A Monoclonal Antibody Against Human Pancreatic Cancer

Objective:To prepare monoclonal antibody against human pancreatic cancer and identify its characterization ,so as to help clical treatment and eatly diagnosis.Methods: Balb/c mice was immunized with human pancreatic cancer cells,the immunized spleen cells and the Sp2/0 myeloma cells were conventionally fused,The MAbs were screened by indrect ELISA and the tissue specificity of MAbs was identified by flow cytometry and immunohistochemical ABC staining, we estallished the nude mouse model of pancreas carcinoma with human pancreatic cancer cells line 8988. Make use of the antibody to make experiment of radioimmunoimage. Results: we obtained a murine McAbSZ121 belonged to IgG1 subclass,The titer Of McAb SZ121 in ascites was 5 10-5. Showed that McAb SZ121 could react selectively to pancreatic cancer and not recognize other cancer, The antigen recognized by McAb Szl21 was proved to be a protein with a Molecnler of 66000 by western blot. Clear tumor imaging was obtained 4 hours after injection of 99mTc-SZ-121 . the radioactivity in tuomr and theradioactivity ratio of tumor to non-tumor tissue increased as time prolonged. Coclusion:McAb was identified by flow cytometry and immunohistochemical ABC staining,Showed that McAb SZ121 could react selectively to pancreatic cancer, Image of 99mTo labeled anti-pancreatic cancer monoclonal antibody SZ-121 in human psincreas carcinoma xenografts showed that 99mTc-SZ-121 perhaps has some value in tuomr diagnosis and treatment.
