The Study On The Isolation And Purifition About The Anticoagulation Of Bombyx Batryticatus

We found the Bombyx Batryticatus-a Chinese drug has strongactivity of anticoagulation in and out of the body. Initial experiments revealed that its crude extract can lengthen the blood clotting time of mammal significantly. The evidence also showed that this activity caused by a special inhibitor of effxcted on thrombin and fibrinogen gene.For the sake of extracting and purifing the anticongulation in Bombyx Batryticatus, We studied techniques of extracted by decocting in water Which was screened by orthogonal design at first, taking the content of protein in Bombyx Batryticatus as evaluation index, we measurated it by the ultraviolet spectrometry, The content of protein in Bombyx Batryticatus was the highest in the best technical of extract by regurgitating, that js: shattered Bombyx Batryticatus was extracted by water with eight times of dosage of material drug regurgitated 30minutes twice.Second, we developed a purification by gel filtration chromatography and ion exchange chromatography, the result is:we had 4 group by gel filtration chromatography ,in which the third group has activity significiently(P<0.01).we isolatied the group by exchangechromatography ,and we had single group with activity. The mass spectrometry showed that its molecular weight was 1.2 Kda, smaller than that of hirudin (about 7Kda), we presume that target is a novel anticoagulant polypeptide.
