| Objective: To study the influence of YXL formulated by the main principle of dissolving blood stasis and unblocking meridian on the influence on the content of amino-acid neurotransmitter and their receptors' gene expression in the hippocampus of the the chronic epileptic rats induced by PTZ.Methods: 10 SD rats were selected randomly from experimental rats as a normal control group.The rest SD rats were injected abdominally to induce chronic-kindling models by PTZ 35mg/kg for 28 days,then 30 having build-up model rats were divided randomly completely into three groups: model, YXL, VPA.After drug-intervertion, epileptic rat's behaviour, EEG were compared, the amino-acid neurotransmitter in the hippocampus was detected by High-performance liquid chromatography(HPLC),the NMDAR1 mRNA, mGluRl mRNA and GABAAR mRNA gene expression was detected by situs hybridization, then observe posotive-cells shape anddistribute, measuring their mean gray.Resaults: 1. YXL can effectively prolong the potential time and shorten last-time of epilepsy-attacks, decreasing the frequency and amplitudes of waves of epileptic discharge in rats' brain and there was significant diffirence compared with model group(P<0.05,P<0.01).It showed intimate-depended effect between YXL and epilepsy-attack. 2.YXL increases the content of IAAs(GABA, Gly) and decrease the content of EAAs(Glu, Asp) in model rats' hip-pocampus and there was also significant difFirence compared with model group(P<0.05,P<0.01).3. Be compared the model group, YXL can decrease NMDAR1 mRNA and mGluRl mRNA gene expression and increase GABAA mRNA gene expression by detecting mean gray by situs hybridization (P<0.05).Conclusions: YXL can significantly improve epileptic rat's behaviour and EBQ adjusting two kinds amino-acid neurotransmitter and regulating their receptors' gene expression in the hippocampus. We can guess it may be one of the effective mechanisms that YXL can cure epilepsy. |
