Radix Polygonatum odorati, the dry root of Polygonatum odoratum (Mill.) Druce, is a well-known Traditional Chinese Medicine named Yuzhu and belongs to the Polygonati genus of Liliaceae. The main constituents include polysaccharide, saponin, flavonoid, alkaloid, sterone, tannide, phlegm, cardiac glycoside etc. Polygonatum odoratum has been extensively used in treatments for many kinds of diseases such as hypertension. The major constituents of P. odoratum included polysaccharide, saponin, flavonoid, alkaloid, etc. At present, the standard of quality about Radix Polygonati, the content of polysaccharide is not less than 6.0% in China pharmacopoeia (2000 edition).In this research, HPLC was used to develop fingerprint of MeOH extraction of P. odoratum in our studies. One compound was isolated. It was identified by UV, 'H-NMR, 13C-NMR. 13CDEPT135 and MS, named Yuzhu ketone. The content of Yuzhu' acid was determined by using high-performance liquid chromatography. Chromatography conditions were Zorbax XDB-C8 (4.6 mm x 150 mm, 5um) column, The mobile phase were acetonitrile and water, the analyses were performed by gradient elution, and 204nm at detective wavelength, the rate of flow is 1.0 ml/min, the result showed that the retention time of Yuzhu ketone is 24.5min,the content isvery high in Radix Polygonati.To establish fingerprint chromatography of Traditional Chinese Medicine Radix Polygonati by HPLC, The HPLC method was used. Chromatography conditions were Zorbax XDB-C8 (4.6 mm x 150 mm, Sum) column. The mobile phase were acetonitrile and water, the analyses were performed by gradient elution, and 210nm at detective wavelength, the rate of flow is 1.0 ml/min, the result showed that the retention time of Yuzhu' ketone is 54.5min.The result showed that 10 peaks were common. The results of stability, accuracy and reproducibility were accurate. The RSD of relative retention time is not less than 3%. At the same time, Radix Polygonati that produced in different geographical regions and collected in different seasons of harvest were tested by HPLC. The rootlets, stems and leaf were also tested by HPLC; at the same time, Polygonatum odoratum and Polygonatum cyrtonema were tested by HPLC. The method is reliable and provides a reference standard for the quality control of Radix Polygonati.
