Seasickness belongs to motion sickness which is caused by ships. Seasickness may have a great impact on military operations at sea and so the research of anti-seasickness' measures is important topics in civil and foreign military medical sciences. There are transparent disorders in neurological function in seasickness and it is used to be recognized as the result of the brain functional disorder after vestibular stimulation. But the pathogenesis of seasickness is not clarificated, although in more a hundred of years there have been considerable hypothesis, but none of which can best explain the occurrence, development and all kinds of symptoms in seasickness. In all these hypothesis there is one of" themry of the change of blood kinetics", it said that there is tight contact in vestibule and vegetative nerve, so stimulation in vertibule inevitably caused the changes in vegetative nervous system include vascular reactivity. The disorders in vegetative nervous system after seasickness caused many active materials excrete, and some of them are acting on vascular. There have been some researches cued the change of brain blood flow in seasickness. So we choose the rats which in the acute seasickness behavior to research the changes in the supply of blood and oxygen in seasickness stress rat's brain, expect to provide the basis and new think pathway of advanced anti-seasickness research. In our experiment we also choose Chinese traditional medicine which have the effect of expand the blood vessel and observe it's anti-seasickness effects and to explore its probable mechanism.I Methods and contents1 creation of animal models in seasickness stress behaviorSD male rats weight of 200?0g, we use seasickness simulator to proceed rat seasickness, the stimulation is clockwise accelerated from 0 to 120 per second by acceleration of 16 per second, then immediately by deceleration of 45 per second to 0, and at once contra-clockwise accelerated from 0 to 120 by 16 per second ,then decelerated from 45 per second to 0, so repeatedly. In everyday morning we give rats two hours' stimulation. After four days' adaptability breeding, rats were divided into three groups: normal group, seasickness group and drug group. Seasickness and druggroups were all given seasickness stimulation. Three groups were put to death after four days' stimulation.We use the weight of Kaolin to determinate rat's severity degree of seasickness, use the Open-field experiment to determinate if rat in the acute stress seasickness behavior.2 Some substances that can response the changes of brain blood supply inseasickness rats2.1 Changes of vessel active materials in seasickness rats.We adopted radio-immunity method to assay prostaglandin's two kinds of substances: thromboxaneB2 and 6-keto-PGFla in plasma.2.2 Changes of the cortex and hippocampus' electrocardiogram in seasickness rats. Because the directly estimate of brain blood flow is difficult, so we employed EEG toreflect brain blood flow. In this experiment we adopted method of auto-control and embedded chronical electrode, and observed the changes of the cortex and hippocampus' electrocardiogram in seasickness rats.2.3 Changes of the expression of cyclooxygenase-2 in rat's cerebral cortex under different time's seasickness stimulation.We employed the method of reverse-transcription polymerase chain reaction to detect the expression of C0X-2mRNA in rat's cerebral cortex. Because COX is the rate-limiting enzyme in the chemical reaction of arachidonic acid to produce prostaglandin and thromboxane. It has two isomers are COX-1 and COX-2, COX-1 exist in most tissue catalysis to produce PG in maintaining normal physiological function. COX-2 is induced that can't be detected in most tissues. There has been already experiments confirmed that hypoxia can induce the expression of COX-2 in endothelial cell.
