| Objective: To investigate the preventive effect of antioxidant and calcium channel blockade on testicular fibrosis in rats and to explore the ideal drug of preventing testis fibrosis. Methods: Wistar rats were divided into control group, preventive EAO (Experimental autoimmune orchitis) group , EAO model group, high dose , median dose and low dose preventive testicular fibrosis group , testicular fibrosis model group. Testicular fibrosis model established by Tao WANG was applied and made a little change. The sperm number ,rate of malformed sperm,length of testis and the intradiameter of seminiferous tubules were measured. The change of interstitial substance and spermatogenic cell were observed by light microscope and transmission electron microscope. Results: 1, There were no significant differences between the preventive EAO group and the control group (P> 0.05) ,the structure of the preventive group was almost the same as the control group. The sperm number , weight of testis and the intradiameter of seminiferous tubules in EAO group were significantly lower than that of normal group (P<0.05andP<0.01) . The rate of malformed sperm in EAO group was significantly higher than those of normal group (P< 0.01) . The interstitial substance was congested with blood, all kind of components of germ cells dropped in varying degrees, formation of multinuclear germ cells and the invading inflammatory cells were observed under light microscop. 2.. The preventive testicular fibrosis group at high dose and median dose and lower dose were combined into an interfering group. The sperm numberin testicular fibrosis model group was significantly lower than that of interfering group, the rate of malformed sperm in testicular fibrosis model group was significantly higher than that of interfering group. The intradiameter of seminiferous tubules in testicular fibrosis model group was reduced to 53.25% compared with normal group, the intradiameter of seminiferous tubules in interfering group was reduced to 25.39% compared with normal group .Compared among the high dose and median dose and low dose group, the thickness of limiting membrane at high dose and median dose group was almost the same as that of the control group,the damage of spermatogenic cells was lighter than that of the control group. The thickness of limiting membrane of the low group was thicker and the damage of spermatogenic cells was heavier than that of the control group. But the change of the testis structure at low group was still lighter than that of the testicular fibrosis model group. The interstitial substance of fibrotic group was severe hyperplasia ,the fibroblast in interstitial substance was increased.The interstitial mast cells and peritubular mast cells were increased , the thickness of limiting membrane of seminiferous tubules was serious thickened, the damage of spermatogenic cells was severe. Conclusion: 1 , The united use of vitamin E, C and verapamil can significantly attenuate EAO injury. 2, The testicular fibrosis in rats is significantly prevented by vitamin E, C and verapamil. 3, Testicular fibrotic changes may correlate with the increase in the number of testicular mast cells. |
