To study the relationship between S100A4 mRNA expression level and differentiation and metastasis of renal cell carcinoma combined clinical data and pathology together.MethodsThis study was based on tissues from 31 patients with renal cell carcinomas treated surgically from 2001 5th to 2004 5th. After RT-PCR amplification on 31 RCC tissues and paired normal kidney tissues besides the tumor, we detected fluorescence dosage of S100A4 mRNA in tissues, and we obtained S100A4mRNA gene fragments (212bp) .ResultsS100A4mRNA expression was detected in all of the RCC tissues and para-carcinoma tissues using fluorescence. Expression of 29 RCC tissues' S100A4mRNA was higher than paired para-carcinoma tissues. There was 1 case which had both high expression on RCC tissues and para-carcinoma tissues. Of all cases, patients of low-differentiation RCC had higherexpression rate than high-differentiation RCC obviously (7.94 vs 5.06, P<0.001) ; patients of metastasis also had higher expression rate than non-metastasis (9.61 vs 5.53, P<0.001) . There was no clearly difference of expression rate between Granular cell carcinoma and Clear cell carcinoma (6.98 vs 6.02, P>0.05) .Conclusions1. Expression of S100A4mRNA in RCC tissues was higher than in normal kidney tissues besides carcinoma. S100A4 may be one of markers on assessment of patient's condition developing and clinical treatment instruction in RCC.2. Expression of S100A4mRNA in G2-RCC tissues( low-differentiation ) was higher than in Gl-RCC tissues(high-differentiation) . S100A4mRNA was correlated to RCCdifferentiation and may play an important role in prognosis of RCC.3. Differential expression of S100A4mRNA was correlated to metastasis of RCC. Patients of metastasis had a higher expression rate than those of non-metastasis, which provide screening methods for radical nephrectomy with systematic lymphadenctomy.4.
