Objective: Screening the effective antiviral prescription of 4 compound preparations ?Algefacient and anti-inflammatory (AI2, 3, 7, 9μ, on influenza virus(Flu), respiratory syncytial virus(RSV) and coxsackievirus B3 (CVB3) in vitro; and evaluating the anti-influenza virus activity of one effective prescription in mice.Methods: There are 2 stages. In these 2 stages , virazole and antiviral liquor were used as positive control. The 1st stage is to screen the effective antiviral prescription . Here CPE, MTT assay and hemagglitination test were used to evaluate the antiviral activity of AI No,2 ,3, 7, 9 in MDCK and Hep-2 cells. And the Treatment Index (TI=TC50/IC50) was utilized as a comprehensive criterion to assess the effects of the 4 compounds. In the 2nd stage , influenza virus infection model was established in mice, to observe the protection effect to their death , and the inhibitory effect to their pulmonary pathological changes .According to the results of the 1st stage, one effective prescription was selected .3 non-toxic dose(0.1 g 'kg-1 -d-1, 0.5 g kg- 'd-1, 1.2 g kg-1 .d-1) of the selected prescription were given to infected mice for 6 days. And the living time, death rate, lung index, pulmonary virus hemagglitination titer of the mice were utilized for assessment of the anti-influenza activity in vivo.Results:1 AI No,2 3, 7, 9 all can inactivate the biosynthesis of Flu, RSV and CVB3 in MDCK cell and Hep-2 cell. The anti-Flu IC50 were 108.8, 110.8, 117.6, 63.2μg/mL respectively.The anti-RSV IC50 were 95.9, 96.7, 105.2, 66.9μg/mL respectively.The anti-CVB3 IC50 were 101.2, 96.1, 103.7, 67.8μg/mL respectively. But AI No.9 can inactivate Fhu RSV and CVB3 more effectively(P<0.0l). And the antiviral activity of AI No.9 is similar as the virazole and antiviral liquor at the same doseage level(P>0.05).2 According to the TI of the 4 compound, they are sorted into 2 types:(l) high effective(TI>5):AI No.9, virazole and antiviral liquor; (2)effective(20.05), and more effective than 1.5 g ?kg"1 ?d"1 dose of antiviral liquor(P<0.05).Conclusion:1 AI No.9 are found to be the most effective anti-Flu, anti-RSV, anti-CVB3 prescription, thus its respective component can be used to further purify the most active chemical monomers.2 AI No.2 3 7 9 all can inhibit the biosynthesis of Fhu RSV and CVB3 in vitro. But AI No.9 can inhibit Fhu RSV and CVB3 more effectively(PO.Ol). And the antiviral activity of AI No.9 is similar as the virazole and antiviral liquor at the same doseage level(P>0.05).3 0.1 g ?kg"1 ?d"',0.5 g ?kg"1 ?d"],1.2 g ?kg"1 ?d"1 dose of AI No.9 have obvious anti-influenza virus activity in mice (PO.01). The effect of 1.2 g 'kg'1 -d"1 dose is similar to 0.09 g ?kg"1 ?d'1 dose of virazole (P>0.05), and more effective than 1.5 g ?kg"1 ?d"1 dose of antiviral liquor(PO.05).
