Objective:To investigate the effects of IGF-I and/or TNF-α on proliferation,differentiation and apoptosis of C2C12 myoblast.Methods:Murine C2C12 myoblasts were cultured in vitro and were treated with IGF-I and/or TNF-α. The proliferation was measured by MTT, the differentiation was detected by CK activity calculating and the apoptosis was detected by flow cytometry. Results: The proliferation of C2C12 myoblast treated by IGF-I and/or TNF-α is more remarkble than the control within 48h. CK activity of IGF-I group is gradually going up, although the IGF-I,TNF-α and IGF-I+TNF-α groups are all lower than the control. The apoptosis rate in TNF-α group increases significantly compared with the control, but the IGF-I group is lower than control, and the IGF-I+ TNF-α group is lower than the TNF-α group.Conclusion:IGF-I and/or TNF-α stimulate C2C12 myoblast proliferation. TNF-α inhibits C2C12 myoblast differentiation and promotes Apoptosis. IGF-I delays C2C12 myoblast differentiation but protects against TNF-α-induced apoptosis.
