| Objective: To detect the expression of PDGF-B in rat cornea after cautery with alkali and the value of PDGF-B in corneal wound healingMethods: Cornea was wounded by cautery with alkali in sparague dawley rats. PDGF-BB was detected in corneal specimen at different periods by immunohistochemistry and the mRNA of PDGF-B was detected by RT-PCR at different period corneasResult: The result of immunohistochemistry showed that the expression of PDGF-BB is rarely in normal corneas and it is mainly express in the epithelial cells. After alkali burn, the PDGF-BB expression increase and all the epithelial, stroma and endothelial cells express it, and the inflammatory cells and neovascularizations express PDGF-BB also. At about 7 days after corneal alkali burn, the expression of PDGF-BB is the highest. At 30 days after corneal alkali burn, the stroma cells do not express PDGF-BB. The result of RT-PCR showed that the gene expression of PDGF-B increase after corneal alkali burn, after one day of corneal wound, the gene expression is significantly higher than the normal cornea's, from the 7th to the 15th days, the gene expression is the highest and at the 30th day, the quantity of gene expression is not significantly different from the normal cornea'sConclusion: After corneal alkali burn, the gene expression of PDGF-B increase and its quantity is changing during the process of corneal wound healing.
