| Antagonistic Effect of Melatonin on Alzheimer's disease-like cell and Phosphorylated Tau ProteinObjective:To establish an Alzheimer's disease cell model induced by okadaic acid(OA) ,explore the antagonistic effective of melatonin in relation to phosphorylated tau protein .Methods: OA was used to induce the Alzheimer's disease cell model in NG108-15 cell and SH-SY5Y cell. MTT method was used to measure cell viability, immunocytochemical staining and Western blot were used to measure the expression of phosphorylated tau protein. Results: OA can decrease the viability of NG108-15 cell and SH-SY5Y cell with the time and dose,the expression of Ser396 phosphorylated tau protein was increased at 3h,peaked at 6h,decreased at 12h. The expression of total tau protein was similar with Ser396 phosphorylated tau protein.After pretreating with MLT,the lever of phosphorylated tau protein of 6h was decreased and the lever of total tau protein of 6h was increased.Conclusions:1.OA can induce NG108-15 cell and SH-SY5Y cell to AD cell model.2.Melatonin can increase the viability of AD-like cell.3.Melatonin can notably increase the expression of the total tau protein in AD cell model.4.Melatonin has an antagonistic effect on AD cell model and this maybe contributes to the effect of melatonin on phosphorylated tau protein expression of Ser396 . |
