PURPOSES: Build the model of human prostate cancer xenografts in naked mice; To explore the inhibitory effects of Anti-PSCA mAbs in treatments of human prostate cancer xenografts in mice. METHODS: Solid tumor in mice were produced by subcutaneous injection with PC-3 tumor cells in the flanks of the mice. We picked out 15 mice from all of the mice bearing human prostate cancer xenografts. They were divided into two treatment groups(N,; N2 =5) and a control group(N3=5). 200μg Anti-PSCA mAb imported was injected into abdominal cavity of each mouse of the first treatment group. 200pg Anti-PSCA mAb self-prepared and PBS were injected for the second group and the control group respectively. Anti-PSCA mAbs and PBS were administered once every three days for consecutive three times. The mice survival conditions of three groups were recorded during 5 weeks. The PSA in semm, the tumor weights and dimensions of survived mice were measured. The tumor volume inhibition rate was calculated. T-test was performed to compare differences of PSA in serum, tumor weights and volumes between the treatment and control groups. Routine pathological slides of tumor tissue were observed under light microscope to evaluate the range of tumor tissues damaged by Anti-PSCA mAbs. Ultrastructure was observed with transmission electron microscope. RESULTS: These naked mice all survived the injection. The average PSA level in serum of these three groups were (3.28 + 0.55) ng/ml, (3.82 + 0.59) ng/ml and (7.26 + 0.43) ng/ml. The averageweights of tumors of the three groups were (0.95 + 0.17) g, (1.06 + 0.09) gand (3.08 + 0.18) g. The average volumes of them were (164.59 + 14.08 ) mm3, ( 192.85 + 8.65 ) mm3 and ( 548.49 + 19.79 ) mm3. There are remarkable differences between the treatment groups and the control group ( P<0.05 ) .The tumor volume inhibition rate of the treatment group with Anti-PSCA mAb self-prepared is 65.84%. Pathological examination showed coagulative necrosis and and plenty of inflammtory cells infiltrated in tumor tissues Apoptosis was observed by transmission electron microscope. CONCLUSIONS: We build the model of human prostate cancer xenografts in naked mice; Anti-PSCA mAbs is remarkable inhibition to human prostate cancer in mice. |
