PDGF is the main cellular mitogen in serum ,which is an important growth factor in inducing vascular smooth muscle cell proliferation and migration . PDGF is closely associated with atherosclerosis.The expression of PDGF is strictly controlled, which is expressed very lowly or even can not be detected in normal cell. PDGF-A is expressed largely in aeota smooth muscular cell of atherosclerosis.The aim of this experiment is to study PDGF. PDGF-A has been indicated to be an important growth factor in inducing vascular smooth muscle cell proliferation and migration in atherosclerosis. The present study was designed to assess effects of amlopine on levels of PDGF-A mRNA in aortic wall of rabbit model, hence the mechanism of amlopine 's inhibition effects on SMC proliferation and migration, and its antiatherosclerogenesis activity.In this study,40 rabbits were randomized into 4 groups: the control group, the atherosclerosis group, the amlopine-treated group, and the selenium + Vitamine E-treated group. Total RNA was prepared by the guanidimum isothiocyanate/caesium chloride extraction from the rabbit aeota SMC in each group, and the PDGF-A mRNA were detected with the use of dot blothybridization and Northern blot hybridization analysis .Results are as follows:1 In atherosclerosis group it is very high (with the strongest signal);2 In amlopine-treated group, the aorta expressed only 1/200 PDGF-A of that of the atherosclerosis group (with weak signal);3 In selenium + Vitamine E group, the PDGF-A mRNA was also decreased by 30 times compared with the atherosclerosis group(with strong signal).4 PDGF-A mRNA expression level was very low and can hardly be detected by dot blot hybridization in control group; Only one specific hybridization band was seen in Northern blot hybridization, indicating high specificity of our DNA probe.Conclusions are as follows:1 . PDGF-A mRNA can hardly be detected in rabbit aeota SMC in control group.2 .In atherosclerosis group PDGF-A is very high , which indicts that PDGF-A is closely related to atherosclerosis.3 .Amlopine can decrease the level of PDGF-A gene expression.4 The effect of selenium + Vitamine E group on PDGF-A gene expression.is smaller than amlopine...
