Objective:Diabetes Mellitus (DM) is an endocrine metabolism disease and its incidence is high, it is difficult to cure, and it is with multi-complication. Now, DM has been the third serious disease only behind with tumor and cardiovasvular disease. DM does harm to human health. About 65-70% DM patients died of heart diease. Lots of research indicating: metabolic disorder of cadiocyte: accommodation abnormality of intracellular Ca2+ and free radical lead to the damage of diabetic myocardium. Of the total, the relationship between oxidative stress (OS) and diabetic cardiomyopathy is a hot spot. Hyperglycemia can lead to accumulation of a great quantity free radical in myocardium, including reactive oxygen species (ROS) and reactive nitrogen specie (RNS). In these, nitric oxide( NO) reacts with superoxide anion into peroxynitrite (ONOO-), which is a powerful oxidizing agent and the main factor to the damage of oxidative stress. However, if does it has relationship between change of content of ONOO-and myocardial damage in DM? If is ONOO-the main factor to myocardial damage in DM ? if dose Promoting Blood Flow Nourishing yin Recipe (PBFNR) prevents and cures myocardial damage by affacting on ONOO-? It is not yet clear now. According to many dates, the content of TNF-αand IL-1βincrease in not only myocardium but blood plasm. TNF-αand IL-1βcan induce the expressiong of iNOS. If it is induced, its activity can be kept for 20h. And the concentration of the iNOS will be the 1000 times of the concentration synthesized by cNOS. iNOS is mainly responsible for excessive NO. In vivo it also produces O2-.NO reacts quickly with O2-into ONOO-competing with SOD. In this expreriment, the mechanism of the myocardial damage in DM is sdudied through the way of TNF-αand IL-1β-iNOS-ONOO-way. Traditional medicine considers yin asthenia is a key of DM. Yin asthenia lead to DM, whict is the main model-base to most doctors; DM can result in stagnat blood and then induces DM, too.DM is asthenia in origin and asthenia in superficiality and inclusion bewteen asthenia and sthenia. The foundation is deficiency of both vital energy and yin, and the appearance is the blockage of arteries and veins. According to above, Promoting Blood Flow Nourishing yin Recipe (PBFNR) is used to prevent and cure the myocardial damage in DM. Aminoguanidine(AG) is used in control group(It is the selective inhibitor of iNOS and the inhibiting activity is higher 50 times and 500 times respectively than nNOS and eNOS ). Six months Sprague Dawley (SD) male rats are used to duplicateDM model with streptozotocin (STZ). Observing if ONOO-participate the progress of the myocardial damage and approaching the mechanism of PBFNR prevention and cure the myocardial damage in DM. Method 1 rats divided into groups and drawing the materials In our experiment, 37 six-month SD male rats, weight (275.72 ±18.31)g, are divided into four groups randomly, normal control group(8), modellogy group (9) AG group(8), PBFNR group(12). The later three groups are injected at one time into abdominal with STZ solution (40mg/kg weight, dissolved with 0.1mol/L sodium citrate/citric acid buffer solution. PH=4.4 fresh disposal, with concentration of 10mg/ml), and make into diabetic modal (blood sugar higher than 13mmol/L, urine sugar +++ and above). For thirteen weeks after injection, blood sugar, urine sugar and body weight are detected respectively. After thirteen weeks, collecting blood plasm for testing IL -1βand TNF-α. quickly taking myocardium of left free ventricular wall, one part immediately put into liquid nitrogen and then conserving in 70℃for RNA; another part for testing SOD,MDA,NOS and iNOS.the other part for observing morphology. 2 observing of morphology with light microscope and electron microscope of myocardium. 3 testing the content of ONOO-in myocardiumwith immunohistochemistry stain of NT. 4 testing NOS and iNOS in myocardium 4.1 the gene expression of iNOS 4.1.1 extrating of total-RNA of myocardium:with guanidium thiocyanate-phenol-chloroform method of Chomcz...
