Objective :To study the mechnism of transient rise of introculer pressure (IOP) after laser iridectomy. Methods: Changes in the IOP , the activity of superoxide dismutase(SOD),the concentrition of nitric oxide(NO),nitric oxide synthase (Nos), 6-keto-prostaglandinF1α(6-Keto-PGF1α),superoxide dismutase(SOD),Malondialdehyde (MDA) in aqueous 15minutes, 1hour, 6hours, 12hours, 24hours after combined Krypton (wavelength-green:520nm and yellow: 568.2nm, 200um spot size,exposure time of 0.2s and the energy of 300mw) and Q-switched Nd:YAG(the single pulse energy of 10mJ,1pulse) laser iridoctomy were observed.Kr. laser was the first used to create partial iridectomy without penetrating pigmented epithelium,and then Nd:YAG laser was required to complete a patent iridectomy.Iridoctomy were performed in 96 eyes of 48 pigmented rabbits.48 rabbits were choiced by the double blind to get rid of those IOP were high or too low.48 rabbits were divided into six groups at random ,each group had 8 rabbits.The first group which was not performed iridoctomy was the control,The second group was 15 minutes after combined laser iridoctomy.The third group was 1 hour after iridoctomy.The fourth group was 6 hours after iridoctomy.The fifth group was 12 hours after iridoctomy.The sixth group was 24 hours after iridoctomy.NO in aqueous was measured with Griess reaction. Nos was measured with Spectrophotometry.The activities of SOD was measured by xanthine oxidase and MDA by the method of TBA microdetermination.6-Keto-PGF1αwas measured by means of radio immunoassay. Results: 1.The change of IOP The IOP significantly increase at 15 minutes after LI, reach the peak at 001 hour after LI and decrease to the normal level at 12 hours. The IOP of normal group was 21.634±3.0262mmHg, 32.681 ±4.2193mmHg at 15 minutes after LI (P<0.05), 35.699±6.4709mmHg at 1 hour after LI (P<0.05), 26.368±3.5903mmHg at 6 hours after LI (P<0.05), 18.943±2.9982mmHg at 12 hours after LI (P > 0.05), 20.937±4.6934mmHg at 24 hours after LI (P>0.05) . 2. The concentrition of NO The level of NO of normal group was 37.3 ±17.591umol/L, 19.926±5.1935umol/L at 15 minutes after LI(P<0.05) , 16.69±3.9715umol/L at 1 hour after LI(P<0.05) , 22.777±4.5878umol/L at 6 hours after LI(P<0.05) , 39.013±6.7789umol/L at 12 hours after LI(P>0.05) , 36.173±14.1164 umol/L at 24 hours after LI(P>0.05) . 3. The activity of NOSThe activity of NOS in normal group was 16.777±3.8656 U/mgprot, 12.083±1.9732U/mgprot at 15 minutes after LI (P<0.05) , 9.804±1.8493U/mgprot at 1 hour after LI(P<0.05) , 11.234±2.3119U/mgprot at 6 hours after LI(P>0.05) , 13.625±3.5121U/mgprot at 12 hours after LI(P>0.05), 15.646±3.412U/mgprot at 24 hours after LI(P>0.05) . 4. The activity of SOD The activity of SOD in normal group was 113.33±32.1305U/ml, 45.1 ±15.6396 U/ml at 15 minutes after LI(P<0.05), 53.02±22.2024U/ml at 1 hour after LI(P<0.05), 65.96±24.4739U/ml at 6 hours after LI(P<0.05), 76.69±27.2557U/ml at 12 hours after LI(P > 0.05), 99.28 ±40.5519U/ml at 24 hours after LI(P>0.05) . 5. The level of MDA The level of MDA in normal group was 3.0204 ±1.4107nmol/ml, 6.167±1.7493 nmol/ml at 15 minutes after LI(P<0.05), 8.3022±1.7704nmol/ml at 1 hour after LI(P<0.05), 8.6859±1.3827nmol/ml at 6 hours after LI(P<0.05),4.6879±1.5714nmol/ml at 12 hours after LI(P > 0.05), 2.4535 ±0.6882nmol/ml at 24 hours after LI(P>0.05) . 6. The contents of 6-Keto-PGF1αThe contents of 6-Keto-PGF1αin normal group was 144.3 ±56.4388pg/ml, 5395±2151.231pg/ml at 15 minutes after LI (P<0.01) , 4443.8±1749.24pg/ml at 1 hour after LI (P<0.01) , 2241.1±992.1369pg/ml at 6 hours after LI (P<0.01) , 2132±1218.647pg/ml at 12 hours after LI (P<0.01) , 1462.9±...
