OBJECTIVE To study the expression of estrogen receptor α estrogen receptor β progesterone receptor and progesterone receptor B in endometrial carcinoma and the relationship with the clinical-pathological features.METHODS Using semi-quantitive reverse transcription-polymerase chain reaction(RT-PCR), we examined the expression of ER α , ERβ, PR and PRB mRNA and using immunohistochemistry, we examined the expression of their protein in 51 cases of endometrial carcinomas and 47 cases of hyperplasia endometriums as well as 18 cases of normal endometriums.RESULT 1. The expression of ER α and ERβ mRNA (1) (1)ERα mRNA were detected in 72.55% (37/51) endometrial carcinoma, 89.36%(42/47) hyperplasia endometriums and 100%(18/18) normal endometriums tissues.The expression average quantity of ER a mRNA is 0.5279 + 0.1295 in endometrial carcinoma, 0.7329 + 0.1290 in hyperplasia endometrium and 0.6746 + 0.1376 in normal endometriums tissues respectively.The expressions of ER α mRNA in three groups above demonstrated significant difference (P<0.01) .(2)ERP mRNA were detected in 47.06% (24/51) endometrial carcinoma, 82.98%(39/47) hyperplasia endometriums and 88.89%(16/18) normal endometriums tissues.The expression average quantity of ER β mRNA is 0.3025 + 0.1687 in endometrial carcinoma, 0.5458 ±0.1668 in hyperplasia endometriums and 0.4422 + 0.1472 in normal endometriums tissues respectively.The expressions of ER β mRNA in three groups above demonstrated significant difference (P<0.01) . (3)The ratio of the raletive level of ERα and ER β mRNA was lower in endometrial carcinomas than in hyperplasia endometriums and normal edmetriums. (2) CD Among endometrial carcinoma group, lower ER α mRNA expression significantly correlated with pathological grade (P <0.01); the positive rate and the expression of average quantity of ER α were higher in Endometrioid Adenocarcinoma than other histological subtypes (P <0.05); but it has no association with clinical—surgical stage (P>0.05) lymph node metastasis (P >0.05) moymetrial depth(P >0.05) and age(P>0.05 ).(2)LowerER β mRNA expression significantly correlated with histological subtype(.P<0.05). (3) Amonghyperplasia endometriums group, the expression of average quantity of ER αβ mRNA in atypical hyperplasia was significantly lower than in simple hyperplasia and complex hyperplasia tissues; while the expression of average quantity of ER β mRNA had no this change. (4) Among normal endometriums group, the expression of ER α and ER β between hyperplasia stage and secretion stage has no significant difference. (5) The expression of average quantity of ER α mRNA was higher than ER β in normal hyperplasia and carcinoma tissue, respectively.2. The expression of PR and PRB mRNA (1) (1)PR mRNA were detected in 82.35% (42/51) endometrial carcinoma, 91.49%(43/47) hyperplasia endometriums and 100%(18/18) normal endometriums tissues.The expression average quantity of PR mRNA is 0.7251 ± 0.2866 in endometrial carcinoma , 0.9878 ± 0.2148 in hyperplasia endometriums and 1.1004 ± 0.1980 in normal endometriums tissues respectively.The expressions of average quantity of PR mRNA in three groups above demonstrated significant difference (P<0.01) .(2)PRB mRNA were detected in 45.10% (23/51) endometrial carcinoma, 82.98%(39/47) hyperplasia endometriums and 94.44%(17/18) normal endometriums tissues.The expression average quantity of PRB mRNA is 0.3855 ±0.1387 in endometrial carcinoma, 0.5175 ±0.1221 in hyperplasia endometriums and 0.5840 ±0.1299 in normal endometriums tissues respectively.The expressions of PRB mRNA in three groups above demonstrated significant difference(P<0.01 ).(2)(1).Among endometrial carcinoma group, lower PRmRNA expression significantly correlated with pathological grade (P <0.01) and clinical—surgical stage (P <0.01) ; the positive rate and the expression of average quantity of PR were higher in Endometrioid Adenocarcinoma than other histological subtypes (P <0.05); but it has no association with lymph node metastasis (P >0.05) moymetrial depth(P >0.05) and age(P>0.05 ).(2) higher PRB mRNA expression significantly correlated with histological subtype(P<0.05). (3 ) Among hyperplasia endometriums group, the expression of average quantity of PR mRNA in atypical hyperplasia was significantly lower than in simple hyperplasia and complex hyperplasia tissues; while the expression of average quantity of PRB mRNA had no this change. (4) Among normal endometriums group, the expression of PR and PRB mRNA between hyperplasia stage and secretion stage has no significant difference.3. The expression of ER α and ER β protein (1) (1)ER a protein were detected in 66.67% (34/51) endometrial carcinoma, 87.32%(41/47) hyperplasia endometriums and 100%(18/18) normal endometriums tissues.The expressions of ER α protein in threegroups above demonstrated significant difference (P<0.05) .(2)ER αβ protein were detected in 43.14%(23/51) endometrial carcinoma, 78.72%(37/47) hyperplasia endometriums and 83.33%(15/18) normal endometriums tissues. The expressions of ERβ mRNA in three groups above demonstrated significant difference (P<0.05) . (2) (1).Among endometrial carcinoma group , higher ER α protein expression significantly correlated with pathological grade (P <0.01) and histological subtype; but it has no association with clinical—surgical stage (P >0.05 ) lymph node metastasis (P >0.05) moymetrial depth(P >0.05) and age(P>0.05 ).(2)higher ER 3 protein expression significantly correlated with histological subtype(P<0.05). (3 ) Among hyperplasia endometriums group , the expression of average quantity of ER α protein in atypical hyperplasia was significantly lower than in simple hyperplasia and complex hyperplasia tissues; while the expression of ER β protein had no this change. (4) Among normal endometriums group, the expression of ER α in hyperplasia stage was higher than in secretion stage; while the expression of ERβ protein has no significant difference.4.. The expression of PR and PRB protein (1 )(1)PR protein were detected in 76.47% ( 39/51 ) endometrial carcinoma, 87.32%(41/47) hyperplasia endometriums and 94.44%(17/18) normal endometriums tissues.The expressions of PR protein in three groups above demonstrated significant difference (P<0.05) .(2)PRB protein were detected in 41.18%(21/51) endometrial carcinoma, 76.60%(36/47) hyperplasia endometriums and 88.89%(16/18) normal endometriums tissues. The expressions of PRB protein in three groups above demonstrated significant difference (P<0.05) . (2) (1).Among endometrial carcinoma group, lower PR protein expression significantly correlated with pathological grade (P <0.01 ) clinical-surgical stage (P <0.05) and histological subtype (P <0.05); but it has no association with lymph node metastasis (P >0.05) moymetrial depth(p>0.05) and age(P>0.05 ). (2) Lower PRB protein expression significantly correlated with histological subtype(P<0.05).(3)Among hyperplasia endometriums group, the expression of average quantity of PR protein in atypical hyperplasia was significantly lower than in simple hyperplasia and complex hyperplasia tissues; while the expression of PRB protein had no this change.(4)Among normal endometriums group, the expression of PR and PRB protein between hyperplasia stage and secretion stage has no significant difference.CONCLUSION 1. The abnormal expression of ER α and ER 3 was closed related to the development of endometrial carcinoma. Decreased level of ER α might play an important role in the pathogenesis of endometrial carcinoma.
