| Objective To investigate the possibility that Human herpes virus 6 (HHV 6) infection may play a role in the rheumatic arthritis or rheumatoid arthritis(RA) and the association between the HHV 6 active infection and two kinds of disease .Methods HHV-6 DNA from plasma and peripheral blood mononuclear cells was detected with nested PCR in 40 rheumatic arthritis patients,62 RA patients and 138 matched healthy adult donors. Both groups of autoimmune antibody were detected by BN ProSpec as well as the ESR (erythrocyte sedimentation rate in an hour) were assessed.Results ①In the peripheral blood mononuclear cells of 40 rheumatic arthritis patients 21 were HHV 6 DNA positive (positive rate 52.5%), while73 of the 138 controls was HHV 6 DNA positive(positive rate 52.9%). There was no significant difference between rheumatic arthritis patients and controls in the HHV 6 DNA positive rate inthe peripheral blood mononuclear cells (x~2 =0.002,P>0.05); ②In peripheral bloodmononuclear cells of the 62 RA patients 34 were HHV 6 DNA positive (positive rate 54.84%), while 73 were found in control group (positive rate 52.9%). The HHV 6DNA in RA patients showed no difference from the normal controls (x~2 =0.065, P>0.05);There was no significant relationship between the HHV 6 DNA positive rate in PBMCs and SS status in RA patients (P>0.05).③In plasma some of rheumatic arthritis patients and RA patients were HHV 6 DNA positive with nested PCR,while none of the controls was HHV 6 DNA positive, The presence of HHV 6 DNA in plasma represents a unique marker of active infection that can be used to investigate the pathologic effects of HHV 6 in humans; ④ASL , RF,CRP and ESR were assessed by BN ProSpec .ASL increased in patients with rheumatic arthritis ,RF increased in patients with RA.Meanwile CRP increased significant and ESR accelerated in the active patients.⑤ HHV 6 DNA positive rate in PBMCs of patients did not correlate with the positive rate of RF and... |
