Experimental Study On The Effect Of Pubescent Angelica Root And Its Alcohol Extract On Apoptosis Of Brain Cells In Nature Aging Mice

Objective: the real intention of the study researches the effect of Pubescent angelica root and its alcohol extract on apoptosis of brain cells and the ability of learning and memory in nature aging mice. In order to investigate the effective mechanism of the different forms of pubescent angelica root postponing brain aging on the basis of the earlier study results.Method: sixteen-month Kun Ming old 96 mice, the fifty-fifty male and female, were divided randomly into three groups( normal old control group 32 mice, old Pubescent angelica root group and its alcohol extract group). The mice were administrated by Chinese herbs in the course of sixty days. Using the qualified mice to do the water labyrith experiment determined the ability of learning and memory. After tested the mice were chosen 9, 10 and 9 mice of every group in order to do the following experiment. The flow cytometer (FCM) was used to survey brain cell apoptosis of the aging mice and we surveyed the effect of Pubescent angelica root and its alcohol extract on apoptosis of brain cells in nature aging mice. And we observed the shape of brain cells under electronic microscope.Result:(1) The compare of the ability of learning and memory: the numbers of old control group dropped compared with young control group (P<0.01). The numbers of Pubescent angelica root and its alcohol extract apparently increased compared with old control group, and the latter is more apparent. There is significant difference between two. The numbers of its alcohol extract group are more than that of the Pubescent angelica root group (P<0.05).(2) The change of morphology of brain cells: There were the more characteristic early apoptosis under electronic microscope inthe aging groups. The shape of brain cells of Pubescent angelica root and its alcohol extract groups is similar to the normal. (3) The compare of the result of FCM testing: Compared with the young control group, brain cell apoptosis ratio had manifest raising of the old control group (P<0.01). The apoptosis ratio of two treatment groups was apparent loss, compared with the aging control group. There was apparent loss in alcohol extract of Pubescent angelica root (P<0. 01).Conclusion:(1) The ability of learning and memory of nature aging mice remarkably dropped, Pubescent angelica root and its alcohol extract can improve it.(2) The study tests that cell death in form of apoptosis existed in the course of brain aging with the loss of learning and memory capability.(3) Pubescent angelica root and its alcohol extract have the effect of delaying brain aging. However, its alcohol extract is more explicit.
