Purpose To investigate the expression of HSP70 in human diabetic cataract and age-related cataract lens epithelial cells (LECs), clarify the effect of abnormal expression of HSP70 on the formation of human cataract.Methods To make the peplos of human lens to be the experimental object, 144 cases of cataract and 20 cases of control in the end. 144 cases of cataract were divided into four groups: human diabetic cataract , cortical cataract, nuclear cataract and posterior subcapsular cataract. Two cases from the same group as one sample by the principle of the same gender and approximate age. The expression of HSP 70 was assayed using RT-PCR(react against duplicating and getting the enzyme chain together) in the epithelial layer of 20 diabetic cataract, 20 cortical cataracts, 17 nuclear cataracts, 15 posterior subcapsular cataracts and 10 normal cases. Regard β-Actin as the confidential reference items to correct, and carry on statistics analysis to the index that is obtained.Results The expression of HSP70 was significantly higher in the LECs of cataract than in the LECs of clear lens (P<0.001). The expression of HSP70 was no significantly different in human diabetic cataract and cortical cataract (P>0.05). The expression of HSP70 in the LECs was significantly different in human diabetic cataract and in nuclear cataracts, posterior subcapsular cataracts (P<0.05). Among the age-related cataract group, the expression of HSP70 in the LECs was significantly different (P<0.05).Conclusions The development of cataract is correlated with over-expression of HSP70, HSP70 has played an important role during the development and formation of human diabetic cataract and age-related cortical cataract, age-related cortical cataract, nuclear cataract and posterior subcapsular cataract may possess a different pathogenesis.
