Objective To investigate the renal protection and mechanism of Chinese CobraVenom(CCV )in renal ischemia/reperfusion injury rat models.Methods 55 rats were randomly divided into three groups; CCV group(25cases),control group(25 cases) and sham operation group(5 cases). 0.1% CCV(0.4mg/Kg)and 1mL saline solution was infused into abdominal cavity 12hbefore ischema in CCV group and control group respectively. CCV group andcontrol group were randomly divided into 5 teams according to the time whenthe rats are sacrificed. Sham operation group only separated double renalarteries. Samples of serum and kidney acquired on the basis of different time.Renal function was measured by Bun and Scr. Complement c3 and c4 wasobserved . Neutrophil infiltration was assessed by myeloperoxidase(MPO).Thekidney samples were examined by HE stain under light microscopy. Theexpression of ICAM-1 was detected by immunohistochemistry(SABC).Results Bun,Scr and MPO activaty rised gradually and reached peak after12h reperfusion in control group. Complement C3 increased at the beginningand decreased in the end. There was obvious difference only between 1h groupand sham operation group(P<0.05).Significant histological damage can be seenunder light microscope.ICAM-1 expressed mainly in tubular epithelium after12h reperfusion. Serum Bun,Scr and MPO activaty decreased significantly inCCV group than control group and meanwhile serum C3 was lower than shamoperation group.However, There was no difference of C4 between CCV groupand control group. expression of ICAM-1 was inhibited greatly and renal lesionwas relatively slight in CCV group.Conclusion Renal Ischemia/reperfusion induces complement C3 activation andintrarenal deposition of ICAM-1. I Pretreating the rats with ccv can reduce C3 ,ICAM-1 expression and neutrophil influx.
