Expressions Of IGF-Ⅰ,IGF-Ⅱ And IGF-Ⅰ R In Human Prostatic Hyperplasitic Tissues Influenced By 5α-reductase Inhibitor

Objective To investigate the expressions of insulin-like growth factor-Ⅰ(IGF-Ⅰ), insulin-like growth factor-Ⅱ(IGF-Ⅱ) and insulin-like growth factor-ⅠR (IGF-ⅠR) in human prostatic hyperplasitic tissues influenced by 5α-reductase inhibitor on the level of protein. Methods The expressions of IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR in 20 prostatic hyperplasia , 20 prostatic hyperplasia influenced by 5α-reductase inhibitor were detected by immunohistochemistry. The levels of DHT in blood serum in two groups were determined by immunoradiometric assay. The results were analyzed using Kruskal-wallis Test and Spearman Test and pathological study on the prostatic tissues. Results (1) The positive expressions of IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR in control group of BPH,(+) were 15%(3/20), (++) were 40%(8/20), (+++) were 45%(9/20);in group treated by 5α-reductase inhibitor,(-) were 65%(13/20), (+) were 35%(7/20). (2) The positive expressions of IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR in contrastive group,(+) were 15%(3/20), (++) were 35%(7/20), (+++) were 50%(10/20);in group treated by 5α-reductase inhibitor,(-) were 80%(16/20), (+) were 20%(4/20). (3) The positive expressions of IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR in control group of BPH,(+) were 5%(1/20), (++) were 30%(6/20), (+++) were 65%(13/20);in group treated by 5α-reductase inhibitor,(-) were 55%(11/20), (+) were 15%(3/20), (++) were 20%(4/20), (+++) were 10%(2/20). (4) In control group ,the mean levels of DHT in blood serum before administration was 1.96±0.98 nmol/L, the mean levels of DHT in blood serum before operation was1.97±0.99 nmol/L; in group treated by 5α-reductase inhibitor, the mean levels of DHT in blood serum before administration was 1.98±0.71nmol/L, the mean levels of DHT in blood serum before operation was1.18±0.46 nmol/L. The results were analyzed using Kruskal-wallis Test respectively , the difference of IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR was significant.(P<0.01); the results were analyzed using Spearman Test respectively, the expressions of IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR were positively correlated with DHT. Conclusions (1) 5α-reductase inhibitor can wither the stromal , epidermal tissues of BPH and reduce IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR on the level of protein. (2) To suggest that IGF-Ⅰ, IGF-Ⅱand IGF-ⅠR play an important role in BPH pathogenesis and were closely correlated with DHT level in blood serum.
