AIM: To investigate the effects of simulated weightlessness on cellular morphology, proliferation, cell cycle and apoptosis of human gastric carcinoma cell line SGC-7901 and human gastric normal cell line HFE-145. Methods: A rotating clinostat was used to simulate weightlessness. Image-pro4.5 image analysis system was employed for morphometric study. The expressions of PCNA were examined by immunohistochemical stain. The changes of cell cycle were examined by a cytometer. Apoptosis was measured by TUNEL method. Results: Compared with control, the area of cell line SGC-7901 decreased significantly in all period of time when subjected in simulated weightlessness (P<0.01) , the ratio of long and short diameter increased significantly after 36h simulated weightlessness(P<0.01),but did not change significantly in other period of time. The area of cell line HFE-145 increased significantly after 12h simulated weightlessness (P<0.01), but did not change significantly in other period of time, the ratio of long and short diameter did not change significantly in all period of time.Compared with control, the expression of PCNA of cell line SGC-7901 did not change significantly after 12h, 24h, and 36h simulated weightlessness, but decreased significantly in 48h and 72h groups(P0.05 and PO.01). The expression of PCNA of cell line HFE-145 did not change significantly after 24h, 36h, 48h and 72h simulated weightlessness, but decreased significantly in 12h group(P0.05).Compared with control, The cell cycles of cell line SGC-7901 in 12h, 24h, and 36h groups did not change significantly, but the cells in G0+G1 phase in 48h and 72h groups increased significantly(P<0.01 and P<0.05), accordingly, the cells in S+G2+M phase decreased significantly(P<0.01 and P<0.05).
