ObjectiveEndometrial carcinoma is one of the three common malignant tumors of female genital tract. It is about 7 percent of all the female carcinoma. Recently, its incidence is gradually elevated all around the world. Endometrial carcinoma becomes a major disease that threatens female life. Thus, studies on etiology, methods of early diagnosis and effective treatment for endometrial carcinoma become a focus to be investigated. More and more studies have demonstrated that tumor angiogenesis plays an important role in tumor growth and metastasis . A new en-dothelial cell marker,CD105,highly expresses on the surface of endothelial cells in tumor tissues with angiogenesis . 80 per cent of endometrial carcinoma is en-dometrioid adenocarcinoma. The purpose of our study is to investigate the correlation between angiogenesis and the growth , metastasis and prognosis of tumor by detecting the microvessel density ( MVD) of endometrial carcinoma. To study the expression of PCNA and the relationship of CD 105 and PCNA in endometrial carcinoma. And the more important thing is that it gives us a new idea in anti-cancer treatment by inhibiting the angiogenesis.Materials and Methods1. Materials77 samples of clinical specimens were abtained from Department of Pathology , the second Clinical College of China Medical University , during the periodfrom June of 2000 to November of 2004 , including 56 endometrioid adenocarcinoma ,21 nonnal endometrium. The age of patients ranges from 35 to 71 years old,with a mean age 53.2 .In endometrioid adenocarcinoma ,surgical stage according to FIGO (2000) :42 of I and II ,14 of 1 and IV;histological grade: 15 of low differentiation,21 of middle differentiation and 20 of high differentiation. In normal endometrium ,10 of prolifertive, 11 of secretory. All the samples are diagnosed by pathological examina tion.2. Main reagentsMouse anti -human CD 105 monoclonal antibody; mouse anti -human PCNA monoclonal antibody, etc.3. MethodsExpression of CD105 and PCNA in the specimens of 56 cases with endometrioid adenocarcinoma and 21 cases with normal endometrium was investigated by sp immunohistochemistry.4. Statistical analysisTake statistical analysis with software of SPSS 11.0.Experimental results1. The expression of CD105 in the different endometrium tissues.CD 105 was expressed in normal endometrium tissues and endometrioid adenocarcinoma tissues,but expression of CD105 was found to be significantly higher in endometrioid adenocarcinoma tissue than in the normal endometrium tissue (p<0.05).2. The relationship between expression of CD 105 and clinical pathological feature of endometrioid adenoc arcinoma .Expression of CD105 in endometrioid adenocarcinoma was closely associated with the clinical stages and lymph nodes metastasis (p <0.05) , but not with differentiated degree ( p >0.05).3. The expression of CD 105 , PCNA in endometrioid adenocarcinomaWith the increase of the expression PCNA in endometrioid adenocarcinoma, the expression of CD 105 also increases (p <0. 05).
