ObjectivePass to study the immunohistochemistry of different grades Astrocytic Tumors, evaluate the significance of CD - 105 expression in human Astrocytic Tumors.MethodsPatient selectionTissue samples ofc ( n = 10) were obtained from patients who underwent surgery in the Second affiliated hospital of China Medical University between A-pril,1999 and March,2004. All tumors were graded according to Word Heath Organization criteria( WHO grade 1 - 2 n = 27, grade 3 n = 14, grade 4 n = 1).ImmunohistochemistryPositive signals were detected by the S - P method with UltrasensitiveTMS -Pkit. Sections were also counterstained with hematoxylin. The labeling indexs of CD 105 were calculated microscopically field at a magnification of 400 by counting at least 1000 tumor cells form selected fields.Result determination: Microvessels density ( MVD ) was calculated in CD105. First, select a microscopically field at magnification of 40 x , search the hot - spots of microvessels, then calculated at a magnification of 400 X by 3 selected. ≤1 MVD : (-), 12 test was used to compare the expression of CD 105 in astrocytic tumors and non - tumor brain tissues. Non - parametic Spearman rank correlation coefficients were used to assess the degree of association among the histopathologic grades of tumors,CD105. Statistical significance was defined as P <0.05.ResultThere are no expressions in natural brain tissues endothelial cell,but there are high expressions in Astrocytic Tumors. (Astrocytic Tumors I - H 37.0% , 10/27;Astrocytic Tumors III - IV-.73. 3%,11/15. There is not a significant difference between the grads I and II. It proved that CD 105 is only expression in the EC of tumor, and there is no expresson in natural EC. The expressions of CD105 in the low malignant Astrocytic Tumors is negative,and the expressions of CD 105 in the high malignant Astrocytic Tumors is positive. Nonparametric Spearman rank correlation analysis identified that the tumor grade was positively correlated with CD 105 expression. DiscussionCD 105 is one of the cell membrance - need proteins. 1985, Quackenbush obtained that Mab have 39 different grads member - antibodies , which is loading in the surface of non T non B acute lymphphoblastic leukemiaby HOON. One of them is mainly loaded in EC, is CD105. 1993, Fernandez Ruiz found that CD105 was loaded in 9q34 qter.CD105 is a new sign of blood vessel. The study of Bourdeau said that the mutation of CD105 will cause the hereditary hemorrhagic telangiectasia; and it was confirmated by the observing Endoglin - deficient mice. Li found that CD105 antagonizes was the inhibitory signaling of transforming growth factor be-tal on human vascular endothelial cell. So CD105 is one of the strong expressions sign on the new endodermis cell. Miller found that Elevated expression of endoglin was a component of the TGF - beta - receptor complex, correlates with proliferation of tumor endothelial cells. Miller signed the endothelial cell of lung cancer tissues and natural lung tissues in CD34.
