Objective To investigate the anticancer activity of taurine against breast cancer induced by 7,12-Dimethylbenz(a)anthracene(DMBA)in rats, and the effect of taurine supplementation on immune, antioxidative activities.Methods 1. Animal test: 77 female Wister rats, aged 7 weeks, were randomly divided into five groups based on body weight. The rats were given 0.5%, 1.0% and 1.5% taurine in group A, B, C, respectively; and free from taurine in group D as the control, while group E was tumor control. After supplemented with taurine one week, rats in A, B, C and E were given 10mg/100g DMBA by injection once for inducing growth of breast cancer. The period of the trail was 18 weeks. 6 weeks later the rats were palpated once weekly to determine the size, location of each tumor. T-cell proliferation and serum SOD, MDA, GSH-Px, O~6-MeG were measured. 2. Cell culture: Hela cells were exposed to various concentrations (200 μ g/ml , 100 μ g/ml , 50 μ g/ml) of taurine for 1 to 3 days. Its proliferations were evaluated by MTT assay.Results 1. Animal test: The tumor incidence of rats (82.35%) in group E was higher than those in group A (53.33%), group B(46.67%) and group C(40.00%) (p<0.05). There was statistically significant difference in mean latent periods of tumor among four groups. It was shortest in group E (80.64d) and longest in group C (93.86d, P<0.01). Mean tumor weight in rats in A, B and C groups supplemented with taurine was lower than that in group E (P<0.05).The rates of tumor inhibition are 40.86% in groupA, 59.73% in group B and 65.82% in group C.2. Mechanisms: The rates of blood T-cell proliferation and spleen T-cell proliferation indexes in taurine supplemented groups were higher than that in group E. Compared with group E, serum SOD activities were increased and MDA were significantly decreased in taurine groups (P<0.01). There were no significant differences of GSH-Px activity between taurine supplemented groups and control groups. The decrease of serum O~6-MeG was observed in taurine supplemented group.3. Cell culture: A decrese of the proliferation was found in Hela cells treated with taurine, The OD value of which was lower than that of control group (p<0.05) after supplemented taurine 48 hours.Conclusion The results suggested that taurine-supplemented diet could effectively inhibit the growth breast cancer induced by DMBA in rats and proliferation of Hela cells in vitro. The antioxidation activity, immunoregulation, DNA repairing and proliferation inhibition might be possible mechanisms of taurine against the tumor.
