Purpose To investigate the clinical manifestations and changes of tumor necrosis facter(TNF-α) in the aqueous humor of the experimental rabbit eyes after injection of lipopolysaccharide(LPS) in different sites and concentrations.Materials and Methods LPS was injected into the subconjunctiva, anterior chamber and vitreous body, clinical manifestations was observed by slip lamp every 4 hours. Rabbits was killed 24 hours after LPS challenge in rabbits .aqueous humor was sampled to examine the cell count, protein content and TNF-α, Histologic grading of inflammation was performed on hematoxylin-eosin-stained sagittal sections of enucleated eyes.Results Different clinical signs was observed, 84.79±6.25pg/ml TNF-α was detected in the aqueous humor after the injection of 10 microgram lipopolysaccharide into the anterior chamber and 143.42±9.31pg/ml when 20 microgram lipopolysaccharide was injected, cell count,protein content and histologic grading varied with different sites injection and doses, and the difference between the test and the contol group was statistically significant. 74.73±7.24pg/ml TNF-α was detected in the aqueous humor after 0.5 microgram lipopolysaccharide was injected into the vitreous body, cell count and protein content in the aqueous increased significantly compared to the control groups. While TNF-α was not detected in the eyes that LPS given subconjunctival, and the inflammation was not statistically significant compared to the control group.Conclusion TNF-α was expressed in the aqueous humor after LPS given intracamerally and intravitreally, with severe clinical and histologic grade of inflammation in the rabbit eyes. While injection into the subconjunctiva didn't cause the expression of TNF-α in the aqueous humor.
