| Objective To observe effects of risedronate on the expression of tumor necrosis factor alpha (TNFa) and osteoprotegerin and alkaline phosphatase activity in human osteoblastic-like cell line MG63 cells and to study the effects of risedronate on the function of osteoblast.Methods Cultured human osteoblastic-like cell line MG63 cell was stained with Van Gieson procedure and modified Gomori's assay on the 6th day, 12th day, 18th day.Mineralized matrix staining was performed with 0.1% paraformaldehyde on the 18th day. MG63 cells were treated with different concentrations of risedronate (10-10mol/l;10-9mol/l, 10-8mol/l 10-7mol/l)for 72 h,and enzyme-linked immunosorbentassay was performed to detect the change of the expression of TNFa and OPG in conditioned media; alkaline phosphatase activity in conditioned media was measured using colorimetric method .Results Type 1 collagen,alkaline phosphatase staining were positive since the 6th day;on the 18th day, type 1 collagen,alkaline phosphatase staining were strongly positive in human osteoblastic-like cell line MG63 cells;on the 18th day, mineralized matrix nodule can be observed. OPG in conditioned media was dose-dependently upregulated by risedronate, TNFa in conditioned media was dose-dependently down-regulated by risedronate.ALP activity was also increased after risedronate treatment.Conclusion Risedronate may inhibit osteoclastogenesis through up-regulating the expression of OPG,down-regulating the expression of TNFa in human osteoblast-like cell MG63. ALP activity was also increased after risedronate treatment. Risedronate may promote the bone formation. |
