| Objective : To study the dynamic expression of matrixmetalloproteinase2(MMP-2) and tissue inhibitor of metalloproteinase(TIMP-2)in normal, proliferative lung tissue and lung spuamous cellcarcinoma(SCC) tissue and their feasibility of acting as maker to monitorthe oncogenesis, progress, invasion, metastasis and prognosis for lungcancer. Methods :Immunohistochemical SABC method was used to detectthe expression of MMP-2 and TIMP-2 in 60 lung spuamous cell carcinomatissue, 30 proliferative and nontypically proliferative lung tissue and 30normal lung tissue. Results : MMP-2 and TIMP-2 in SCC mainly express inendochylema of tumor cells.The positive expression rate of MMP-2 andTIMP-2 in SCC is 73.33% (44/60) and 63.33%(38/60) relatively, inproliferative bronchus epithelium mucosae are both 50%, and in nomalbronchus epithelium mucosae is 26.7%(8/30) and 30.0%(9/30) relatively.Significance difference of positive expression rate of MMP-2 (χ2=18.215,P=0.000)and TIMP-2 (χ2=8.943, P=0.011) were found between lungspuamous cell carcinoma tissue and normal lung tissue, proliferative lungtissue, and between proliferative lung tissue and normal lung tissue.Thepositive expression rate between SCC with nodal metastasis group(100%)and non-nodal metastasis group(52.94%) had significant difference(χ2=16.684,P=0.000). Significant difference also was found in differentstage (χ2=13.295,P=0.001) and grade(χ2=9.383,P=0.025) in SCC tissue.Positive expression rate of MMP-2 between survival<3 year group(90.90%)and survival≥3 year group(63.15%) had significance difference(χ2=5.487,P=0.019).Expression rate of MMP-2 and TIMP-2 (rs=0.818,P=0.001), MMP-2 and MMP-2/TIMP-2 (rs=0.885, P=0.001)in normal,proliferative lung tissue and lung spuamous cell carcinoma tissue hadpositive relation. Positive expression of MMP-2 is an independentprognostic factor by Cox regression(P=0.001). The survival of lung cancerpatients with and without positive expression of MMP-2(P=0.001, LogRank test) and TIMP-2 (P=0.001, Log Rank test) had significantdifference. Conclusions :The activation of MMP-2 gene and disequilibrium ofMMP-2/TIMP-2 may be immportant factors for oncogenesis of lung.Thepositive expression of MMP-2 and MMP-2/TIMP-2 could act as areference to evaluate invasion,metastasis and prognosis in lung carcinoma. |
