Purpose: To study the effects of the corneal endothelium of rabbit after beinginjected Lidocaine, Alcaine, Benoxil into the anterior chamber.Methods: Thirty-six adult rabbits were employed. All rabbits were randomlydivided into Lidocaine group, Alcaine group, Benoxil group and control grouprespectively, nine rabbits(18 eyes) for each group. Six rabbits(12 eyes) wererandomly collected and examined under the contact endothelial specularmicroscope for endothelial cell number, cell density, average cell area,coefficient of variation of cell area(CV) and ratio of hexagonal cells. Threerabbits(6 eyes) were examined under the contact endothelial specularmicroscope for the five quantitative indexes, soon these rabbits were killed, allcorneal endothelium were observed by histopathology and transmission electronmicroscopy at 1 day, 3 days and 14 days after being injected.Results: These finding suggested that Lidocaine, Alcaine, Benoxil can causelesion of corneal endothelium. To Alcaine group, the ultrastructural and themorphology changes would happen in 3 days after injection, but these changeswould almost recover in 14 days after being injected compared with that in thecontrol group; there was not almost changes of the ultrastructural and themorphology for Lidocaine and Benoxil groups after being injected. Endothelialcell number, cell density, average cell area, coefficient of variation of cell area(CV)and ratio of hexagonal cells showed significant difference betweenAlcaine group and control group in 1 day and 3 days after being injected(P<0.05); in 3 days, there was a significant change of the five quantitativeindexes between Lidocaine and Benoxil group with control group(P<0.05).There was no significant difference in 14 days after being injected in each groupand control group(P>0.05) except a significant change of CV between Alcainegroup and control group(P<0.05). One day after being injected, there was asignificant difference of the five quantitative indexes among three experimentalgroup(P<0.05), in 3 days, in 14 days after being injected, there was nosignificant difference of these indexes among these groups(P>0.05).Conclusion: Lidocaine, Alcaine and Benoxil can cause damage of cornealendothelial cells in 3 days after being injected. Furthermore the damage ofAlcaine was higher than that of Lidocaine and Benoxil. But these changes willalmost recover or alleviate in 14 days after being injected.
