Alzheimer disease is a neuro-degenerative disease. The anti-oxidation may be an effective way in the prevention and treatment for AD. Objective: To investigate the probably preventive and therapeutic effect of schisandrone on AD and the mechanism. Methods: In vitro part of our research, we established A6-induced cell model withneuro-damage from primitive-cultured microglia of newborn rats' hippocampus. In vivo, The animal model of AD was established by Aβ stereotactic injection into lateral cerebral ventricle of rats. The intervention of schisandrone was administrated. We observe schisandrone's effects on the expression of IL-1β and iNOS in the hippocampal microglia of rats by RT-PCR, the rats' ability of learning and memory by Y maze. Results: Schisandrone might inhibit the activation and proliferation of microglia, downregulate the transcription of IL-1β and iNOS, and improve the learning and memory ability of rats. Conclusions: Schisandrone could suppress the oxidative stress and inflammatory reaction induced by AB which suggested a protection from AD.
