Objective: To establish a model of osteoblast cultured in vitro and to observe and investigate the effect of dipsacus asperoids on osteoblastic proliferation and differentiation. Methods: human trabecular bone of ilium was obtained to osteoblast culture , then ALP dying and HE dying were applied to identify the osteoblasts. Cells of successive transfer culture were seeded by 5×104/ml and cultured in 24 well cell culture cluster. After confluenced, the extracts of dipsacus asperoids was applied to cell culture, then the ALP content in the culture fluid was detected. Cells were seeded by 2×104/ml in 96 well cell culture cluster. Afer cultured by the extracts of dipsacus asperoids, the function of osteoblastic proliferation was detected by MTT method. Results: the extracts of dipsacus asperoids with different concentration(1ug/ml, 10ug/mk, 100ug/ml) can increase the ability of osteoblastic proliferation; the extracts of dipsacus asperoids with concentration of 10ug/ml and 100ug/ml can increase the ability of osteoblastic differentiation. Conclusion: the extracts of dipsacus asperoids with certain concentration can improve the osteoblastic proliferation and differentiation.
