| Objective To investigate the effects of genistein on bone mineral density(BMD) and bone morphometry in orchidectomized rats. Methods 40 male Sprague-dawley rats,3 to 4 months old,were randomly divided into five groups: sham-operate(Sham),orchidectomized(Orch),orchidectomized supplied diethyl stilbestrol (DES,5μg/d) or genistein (Genâ… 1mg/d,Genâ…¡2mg/d).After 16 weeks with low calcium feed ,the bone mineral density in lumbar spine and femoral neck for all rats were measured by Hologic QDR-2000+DEXA after sacrifice. The bone histomorphometric parameters of left proximal tibia was analyzed by pathology image analysis systems of DMR+Q550. Result (1)BMD: BMD of lumbar and femur of Orch group was lower than sham group(P<0.05). BMD of lumbar and femur of Orch+DES and Orch+Genâ…¡groups were higher than Orch group(P<0.05). BMD of femur of Orch+Genâ… group was higer than Orch group(P<0.05). (2) Bone hismorphometric parameters: Compared to sham group, TBV/TTV and MTPD were lower and MTPS was higher in Orch group(P<0.05). TBV/TTV and MTPT of Orch+DES group were higher than Orch group(P<0.05). TBV/TTV and MTPT of Orch Genâ… and Orch+Genâ…¡were higher than Orch group(P<0.05),but MTPS of Orch+Genâ…¡was lower than Orch group(P<0.05). Conclusion Genistein(1mg/d ,2mg/d)and diethyl stilbestrol(5μg/d)can reduced bone loss,increase the number of bone trabecula and reduced the separable degree of bone trabecula on orchidectomized rats.The effect of genistein(2mg/d) on bone is more than genistein(1mg/d). |
