| China is rich in natural medicinal herbs, and it is an importantway to find a new medicine from the effective natural herbs withthe advanced chemical isolation technics. Melon belongs to cucurbitfaculty melon category. Melon vine effective part(MVEP) is aneffective part distilled from melon vine.In order to find outwhether MVEP has protective effect on liver and explore it'sfunctioning mechanism, we have done three experiments as follows:1. the protective effect of MVEP on the acute liver injury inmice induced by CCL4.The experimented animals were divided into 6 groups by weightand sex: normal group, CCL4 control group, Huganjiaonang(HGJN)group (1.8g.kg-1), MVEP high (3g.kg-1), mid(1.8g.kg-1), low group(0.6 g.kg-1). The drugs were administered by ig, 6 days in series,on the 6th day, 1h after the administration, 40%CCL4 was given bymouth, 16h later, blood was taken from the back eyepit, separatedthe serum. Result: the GPT and GOT level were obviously increased,each group of MVEP inhibited the increase of GPT and GOT, whichindicated the protective effect of MVEP.2. The protective effect of MVEP on the acute liver injury inrat induced by CCL4In order to further approve the protective effect of MVEP onliver and whether it has effect on the level of TBIL and TP in serum,the second experiment was carried out. The groups were divided inthe same way as before, the dose of each MVEP was 3.5 g.kg-1, 2.1g.kg-1 and 0.7 g.kg-1 , the dose of the HGJN group was the same asthe mid group. On the 1st and the 4th day, except the normal group,each group was sc 25% CCL4 5ml.kg-1. On the 2nd day, the drugs wereadministrated by ig, the normal group and model group were givendistilled water. On the 7th day, rats were kept without food but waterwas given as usual, on the 8th day, blood was taken from abdominalaorta, the serum was separated; the liver was weighed to calculatethe liver quotient; and histopathology in liver tissue was examined.Result: The change of the liver: â… group: the surface of the liverwas smooth, they are brunneus, soft ; â…¡group: the volume of liverwas bigger, the liver is hard and yellow; â…¢,â…£,â…¤,â…¥group: thechange of morphology was between the â… group and â…¡group. Theliver quotient of model group was bigger than the normal group,while the liver quotient in high MVEP dose group was lower than themodel group differentially. Each group of MVEP could check theincrease of the content of GPT,GOT,ALP,TBIL in serum,while theHGJN drug did not have such effect. And the level of TP made nodifference in each group. The result of the histopathology: â… group:the structure of the liver lobule was clear,there was no hepatocytedegeneration,necrosis . â…¡group: the structure of liver lobulewas damaged obviously, there were many necrosis ,and the hepatocytebecame broadly fatty degeneration, and many inflammation cells inthem. â…¢,â…£,â…¤,â…¥group: the degree of hepatocyte degeneration,necrosis were generally relieved.3. the protective effect of MVEP on the chronic liver injury inrat induced by CCL4.In order to explore the functioning mechanism of MVEP on liverprotection, and whether it has effect on the liver fibrosis, chronicliver injury in rat was induced by CCL4. The groups were divided asbefore, and the dose of drugs on each group was reduced to half ofthat applied to the acute liver injury in rats. Each wasadministrated the drug by ig, 1 time each day ,6 weeks in series.On the 2nd day, except the normal group each group was ip 30%CCL42ml.kg-1 , twice a week, 5 weeks in series. After the lastadministration, rats were kept without food but water was given asusual, 24h later, blood was taken from abdominal aorta, the serumwas separated; and the histopathology in liver tissue was observed;we made 10% liver homogenate with PBS; the liver DNA was extractedaccording to the instruction of the reagent box, the agar gelelectrophoresis was made. Result: the high and mid group of MVEPcould restrain the increase of GPT and TBIL. The TP level of modelgroup was decreased dramatically while the level of TP of each groupof MVEP and HGP group was higher than model group. The level of MDAin liver homogenate in model group increased obviously, othergroups could check the level of MDA; the activity of SOD in liverhomogenate in model group was lower than normal group, while thehigh, mid group of MVEP and HGP group increased the activity of SOD.The result of histopathology: â… group: the structure of the liverlobule was clear,there was no hepatocyte degeneration,necrosis. |
