| In this research making the local cerebral ischemia model in rats with use of thermocoagulation.The successful model rats were divided into 5 groups :model group, acupuncture group , local point-blocked group ,normal group and sham-operated group at random.then showing the distribution condition of vasoactive intestinal polypeptide (VIP) on the vessels with the SP method of immunohistochemistry(IHC) within the 3hours,6hours and 24hours different time. At the mean time we evaluated the expression of CGRPmRNA by means of in situ hybridization.The results showed that firstly observing under the optical microscope after dying by HE method ,It has been found that 3hours after MCAO, the vessels in the infarction region and its surrounding area of the cerebral ischemia group show a highly spastic condition, while the acupuncture group show the distension. After 6hours, the most vessels of the cerebral ischemia group show the distension, while the acupuncture group show the contraction. After 24 hours, the vessels of the cerebral ischemia group still show the distension, while the acupuncture group still show the contraction. Secondly after 3, 6 and 24 hours of MCAO, on the display of positive immunity signal of VIP on the vessels in the infarction region and its surrounding area and the amounts of vessels with positive immunity signal of VIP, the acupuncture group is significantly different from cerebral ischemia group in the same phase, and no significant difference between cerebral ischemia group and local point-blocked group in the same phase.Thirdly the expression of CGRPmRNA of model group and local point-blocked group significantly descended for all the phase. And the positive signal of acupuncture group is much stronger than model group(P<0.01).Yet the result of local point-blocked group is similar to the model group. So it shows that puncturing Renzhong point can promote the expressing of CGRPmRNA descending in the actue cerebral ischemia rat. Fourthly the stimulus of puncturing Renzhong point might promote the transmitter release of VIP-ergic nerve fiber in brain through a certain neuro-transmission routes, and increase the expression of CGRPmRNA in the neuron thus giving free rein to the VIP function of dilating... |
