Objective:To explore the interference effect of KanXianZengZhi (KXZZ) capsule on hippocampal formation lesion and the ability of learning and memory in epileptic rat kindled by pentylenetetrazol (PTZ). Methods:PTZ kindling was used to duplicate the epileptic rat model. All rats were randomly divided into six groups: KXZZ high-does group, KXZZ middle-does group, KXZZ low-does group, valproate magnesium group, model group and normal control group. Then the convulsion latency was observed and recorded, the ability of learning and memory was tested by Morris Water Maze (MWM) method and the hippocampal formation lesion was monitored through Timm stain method and electron microscope. Results:1. Compared with model group, the convulsion latency among different therapy groups showed much longer time (p<0.05). KXZZ high-does group and valproate magnesium group had the better effect than other therapy groups.2. In model group, the course of learning was delayed and the ability of learning acquisition and memory retaining was decreased obviously in MWM test. Among three different KXZZ dose groups, especially in middle-dose group, the course of learning was shortened and the ability of learning acquisition and memory retaining was improved markedly.3. (l)Mossy fiber sprouting was obvious in the hippoampal CA3 section and the molecular layer of dentate syrus in model group. The percent of sprouting density in model group was higher than that of normal control group (p<0.05). In KXZZ middle and high dose group, the percent was much lower than model group (p<0.05) but still higher than normal control group (p<0.05). The result of Timm stain method suggested that KXZZ capsule could efficiently deterred mossy fiber sprouting in hippocampal CA3 section and dentate syrus.
