| Objectives: To study the synthesis of Selenium-Chitooligosaccharides, and to explore not only the intervening effects but also the therapeutic mechanism of the rat model of diabetes mellitus, and then to provide a theoretical support for developing Selenium-Chitooligosaccharides as a daily healthful beverage for diabetic patients. Methods : Part1: The synthesizing and the token of Selenium-Chitooligosaccharides:commix chitooligosaccharide with selenium will synthesize Selenium-Chitooligosaccharides complex, and examine the complex with ultra violate & infrared spectrogram, then determine the content of selenium in Selenium-Chitooligosaccharides complex. Part 2:Wistar mice were randomized into two groups with similar body weight: groups A, the animal normal group consisting of 20 wistar mice received cool water and ordinary feedstuffs; the animal model group consisting of 70 wistar mice received high-fat high-sucrose diet and cool water. 4 weeks later, the animal model groups were injected STZ (1%STZ 25mg/kg,ip). Then 1 week after the injection the forty animals with impaired glucose tolerance were randomized into four groups: group B were fed with Selenium-Chitooligosaccharides; group C were fed with selenic acid; group D were fed with Chitooligosaccharides; group E (comparison group) and group A (normal group) were fed with the same volume NS; and all the groups are lavaged with the medicine. The treatment lasted for 20 weeks. Blood samples were withdrawn from the end of tails of the animals for observing the effects of Selenium-Chitooligosaccharides on serum fasting or pasting glucose, T-ch and, SOD, MDA and GSH-Px, and insulin at weeks 0, 4,8,12,16,20.IPGTT and insulin sensitive index(ISI) were taken to evaluate insulin... |
