Objective We measured Lactate dehydrogenase-x (LDH-X) activity in sperms and seminal plasma from infertile men with varicocele, to explore the LDH-X in sperm and seminal plasma; to explore the meaningness of Lactate dehydrogenase-x (LDH-X) activity in sperms and seminal plasma is meaningful to the diagnoses of infertility of men with varicocele.Methods 107 patients were collected in this study, they were all infertile men with a medical history of two years or more. We confirmed the diagnosis of the patients according to the clinical physical examination and the standard of WHO. Thirty-four men with varicocele (group I), Forty-two infertile men without varicocele (group II) and thirty-one men with normal fertility (group III) were investigated. Semen was analyzed according to the standard procedure by computer aided of semen analysis. We measured the quantity of the semen , the sperm concentration ,sperm motility, sperm survival rate and Sperm morphology. We detected the activity of LDH-X using semen lactate dehydrogenase x isoenzyme (LDH-X) quantitative assay kit.Results 1. The difference of sperm survival rate between group III and other two group was significant (p<0.01). 2. The difference of ideal sperm morphology rate between group III and other two group was significant(p<0.01). 3. The difference of the percentage of grade A and grade B sperm between group III and other two groups was significant (p<0.01). 4. The difference of the sperm concentration between groupIll and other two groups was significant (p<0.05). 5. The difference of LDH-X activity in sperm, the difference between group I and group III was significant (p<0.01), the difference between group II and group III was significant (p<0.01).The difference was no significant in LDH-X activity in seminal plasma and LDH-X activity in seminal plasma-in all sperm ratio between three groups.6. LDH-X activity in sperm have positive correlation with sperm survival rate ,the percentage of grade A and grade B sperm, ideal sperm morphology rate. 7. LDH-X activity in seminal plasma have no correlation v/ith sperm survival rate ,the percentage of grade A and grade B sperm, ideal sperm morphology rate. 8. LDH-X activity in seminal plasma have positive correlation with sperm concentration.Conclusion 1. LDH-X activity can reflect the function of spermatogenic epithelium. 2.If infertile men with varicocele's LDH-X activity in sperm of was lower than normal, we recommend them to have clinical therapy. 3.The LDH-X in sperm is more valuable than LDH-X activity in semen plasma to the clinical use.
